Due to the fact that synthetic amides are still mainly produced by the aid of coupling reagents with poor atom-economy, the direct catalytic formation of amides from carboxylic acids and amines has become a field of emerging importance. A general, efficient and selective catalytic method for this transformation would meet well with the increasing demands for green chemistry procedures. This review covers catalytic and synthetically relevant methods for direct condensation of carboxylic acids and amines. A comprehensive overview of homogeneous and heterogeneous catalytic methods is presented, covering biocatalysts, Lewis acid catalysts based on boron and metals as well an assortment of other types of catalysts.

Oqtans integrates, for the first time, an assortment of sophisticated machine learning-powered tools into Galaxy, that show superior or equal performance to state-of-the-art tools. Implemented tools comprise of a complete transcriptome analysis workflow: short-read alignment, transcript identification/quantification, and differential expression analysis. Moreover, Oqtans is scalable in the cloud in terms of data storage and computing time needs. Finally, Oqtans and Galaxy facilitate persistent storage, data exchange, and documentation of intermediate results and analysis workflows. We illustrate how Oqtans aids the interpretation of data from different experiments in easy to understand use cases. Users can easily create their own workflows and extend Oqtans by integrating specific tools. Oqtans is available as (a) a cloud machine image with a demo instance at cloud.oqtans.org, (b) a public Galaxy instance at galaxy.cbio.mskcc.org, (c) a git repository containing all installed software at oqtans.org/git most of which is also avaliable from (d) the Galaxy Toolshed and (e) a share string to use along with Galaxy CloudMan.

NMO had distinctive clinical, radiologic, and pathologic characteristics and, in most cases, a highly specific biomarker, aquaporin-4 (AQP4) autoantibodies, was detectable.1 However, recent observations might be interpreted as dashing hopes of NMO being a single disorder with uniform pathogenesis. The clinical presentation of NMO is increasingly recognized as heterogeneous. A variety of cerebral, diencephalic, and brainstem presentations1,2 may occur as presenting features or over the course of the disease, which may have been previously considered inconsistent with the diagnosis. However, most of the manifestations of NMO seem to be explained by the distribution and level of expression of AQP4, so the heterogeneity of clinical manifestations does not seriously challenge the hypothesis of uniform pathogenesis.3,4 A more vexing issue has been the 10%-30% of persons who are seronegative for the AQP4 autoantibody despite evaluation with the most sensitive cell-based assays available. Cell-free microRNAs (miRNAs) have been shown to exist stably in a wide spectrum of body fluids and their expression profiles have been shown to reflect an assortment of physiological conditions, underscoring the utility of this new class of molecules to function as noninvasive biomarkers of disease.Content:We summarize information on the known mechanisms of miRNA protection and release into extracellular space and compile the current literature on extracellular miRNAs that have been investigated as biomarkers of 20 different cancers, 11 organ damage conditions, and 10 diverse disease states. We also discuss the various strategies involved in the miRNA biomarker discovery workflow and provide a critical opinion on the impediments faced by this advancing field that need to be overcome in the laboratory.Summary:The field of miRNA-centered diagnostics is still in its infancy, and basic questions with regard to their exact roles of miRNAs in the pathophysiology of diseases and the mechanisms of their release from affected cells into biological fluids are yet to be completely understood. Nevertheless, these noninvasive micromarkers have immense potential in translational medicine not only for use in monitoring the efficacy and safety of therapeutic regimens but to also to guide the diagnoses of diseases, determine risks of developing diseases or conditions, and more importantly, to inform treatment options.
