- Preparation and characterization of nocodazole-loaded solid lipid nanoparticles.

Moreover, it has low drug release time and high particle size. Solid Lipid Nanoparticles (SLNs) have been evaluated for decrease in particle size and therefore increase in drug release time, for such drugs. In this study, NCD has been successfully incorporated into SLNs systems and remained stable for a period of 90 days. NCD structure related to the chemical nature of solid lipid is a key factor to decide whether anticarcinogenic agent will be incorporated in the long term and for a controlled optimization of active ingredient incorporation and loading, intensive characterization of the physical state of the lipid particles were highly essential. Thus, NMR, FT-IR, DSC (for thermal behavior) analyses were performed and the results did not indicate any problem on stability. Moreover, SLNs were decreased size of NCD in addition to increase in time of the drug release. After SLN preparation, particle size, polydispersity index, electrical conductivity and zeta potential were measured and drug release from NCD-loaded SLNs were performed. These values seem to be of the desired range.

However, their use in food industry is limited mostly because of their poor water-solubility and low thermal stability. Ferritins are widely distributed in nature with a shell-like structure which offers a great opportunity to improve the water-solubility and thermal stability of the carotenoids by encapsulation. In this work, recombinant human H-chain ferritin (rHuHF) was prepared and used to encapsulate β-carotene, a typical compound among carotenoids, by taking advantage of the reversible dissociation and reassembly characteristic of apoferritin in different pH environments. Results from high-performance liquid chromatography (HPLC), UV/Vis spectroscopy and transmission electron microscope (TEM) indicated that β-carotene molecules were successfully encapsulated within protein cages with a β-carotene/protein molar ratio of 12.4-1. Upon such encapsulation, these β-carotene-containing apoferritin nanocomposites were water-soluble. Interestingly, the thermal stability of the β-carotene encapsulated within apoferritin nanocages was markedly improved as compared to free β-carotene. These new properties might be favourable to the utilisation of β-carotene in food industry.

Electronic address: minh.ha@otago.ac.nz.AbstractThe present study investigated the effects of both l- and iso-ascorbic acid (AA) on the activity of four plant proteases (papain, bromelain, actinidin and zingibain) and three microbial proteases (Bacterial Protease G, Fungal 31,000 and Fungal 60,000) preparations using fluorescent-labelled casein, meat myofibrillar and connective tissue extracts to explore their effects on meat structure components upon treatment with individual proteases. While l-AA in the range 0.8-3.2mM inhibited the activity of papain, bromelain and zingibain, iso-AA acted as an inhibitor of papain but as an activator of zingibain and had no significant effect on bromelain. Both AA isoforms acted as an activator of the actinidin protease and the concentration of AA isoforms appeared to affect the level of activation of the protease. The effect of the two AA isoforms on collagen and myofibrillar protein hydrolyzing activity varied depending on the concentration of the two AA isoforms. The results indicate the ability to up and down regulate the activity of the investigated proteases by using an appropriate concentration of the AA isoform.
