Different particle coatings were employed to assess their impact on cellular uptake in vitro. In vivo work was then performed to demonstrate efficacy in tumor-bearing mouse models. Materials and method: Paclitaxel nanosuspensions were prepared using a homogenization process and coated with excipients. Surface charge was measured by zeta potential, potency by high-performance liquid chromatography, and solubility using an in-line UV probe. Cellular uptake studies were performed via flow cytometry. In vivo experiments were performed to determine residence time, maximum tolerated dose, and the efficacy of paclitaxel nanosuspensions (Paclitaxel-NS). Results: A stable paclitaxel nanosuspension was prepared and coated with various excipients. Studies in mice showed that the nanosuspension was well-tolerated and at least as effective as the IV Taxol control in prolonging mouse survival in a head and neck cancer model as well as an ovarian cancer model with a lower overall drug dose than the traditional IV administration route. Conclusions: The paclitaxel nanosuspension is suitable for cellular uptake. The nanosuspension was effective in prolonging life in two separate xenograft orthotopic murine cancer models through two separate routes of administration.

We used two kinds of leukemia cells (K562, U937) as main tumor cell models and cells (peripheral blood mononuclear cells, spleen lymphocytes) separated from healthy ICR mice as normal cell models. The multivolume spectrophotometer system and fluorescence spectrophotometer were used to determine the spectral characteristics of DVDMS. The uptake of DVDMS by tumor cells and normal cells was measured by flow cytometry. The MTT assay was used to examine the cytotoxicity and sonotoxicity of DVDMS. The absorption spectra showed that DVDMS had five distinct peaks at 359, 514, 548, 580, and 631 nm, respectively, and the maximum peak was at ∼359 nm. The fluorescence emission spectra showed that DVDMS fluorescence emission was at 642 nm. DVDMS showed an advantage of quick cellular uptake and selectively accumulated in tumor cells compared with normal healthy cells. The cytotoxicity of DVDMS by the MTT method was dose dependent, and DVDMS had little cytotoxicity to normal cells. The sonotoxicity of DVDMS showed that in the presence of DVDMS, under appropriate conditions, the cell-damaging effect of ultrasound was significantly enhanced. The present study showed that the newly synthesized sensitizer, DVDMS, under appropriate experiment conditions, can act as a potential sonosensitizer for tumors in sonodynamic therapy.

It is thought growth factors may mediate this putative relationship, particularly by augmenting plasticity mechanisms in the hippocampus, a brain region critical for learning and memory. Among these factors, glucocorticoids, brain derived neurotrophic factor (BDNF), insulin-like growth factor-1 (IGF-1), and vascular endothelial growth factor (VEGF), hormones that have considerable and diverse physiological importance, are thought to effect normal and exercise-induced hippocampal plasticity. Despite these predictions, relatively few published human studies have tested hypotheses that relate exercise and fitness to the hippocampus, and none have considered the potential links to all of these hormonal components. Here we present cross-sectional data from a study of recognition memory; serum BDNF, cortisol, IGF-1, and VEGF levels; and aerobic capacity in healthy young adults. We measured circulating levels of these hormones together with performance on a recognition memory task, and a standard graded treadmill test of aerobic fitness. Regression analyses demonstrated BDNF and aerobic fitness predict recognition memory in an interactive manner. In addition, IGF-1 was positively associated with aerobic fitness, but not with recognition memory. Our results may suggest an exercise adaptation-related change in the BDNF dose-response curve that relates to hippocampal memory.
