- The cellulolytic system of Thermobifida fusca.
- Gomez Del Pulgar EM, Saadeddin A.Author information Abengoa Research , Abengoa, Campus Palmas Altas, Seville , Spain.AbstractAbstract The process of bioethanol production from biomass comprises pretreatments and enzyme-mediated hydrolysis to convert lignocellulose into fermentable sugars. Because of the recalcitrant character of cellulose, the enzymatic hydrolysis is considered the major challenge in this process to be economically competitive. These technical difficulties highlight the need for the discovery of new enzymes to optimize and lower the cost of current technologies. Microorganisms have developed efficient systems for cellulose degradation. Among cellulolytic microbes, Thermobifida fusca possesses great physiological and cellulolytic characteristics (thermostability, high activity and tolerance to a broad pH range) making it an interesting organism to be studied from an applied perspective. In this review we describe the main enzymes/proteins produced by T.fusca (cellulases, xylanases, mannanase, manosidase, CBM33 and CelR), the effect of substrate on T. fusca proteome, enzyme improvement approaches, synergism between enzymes/proteins and artificial cellulosomes.

- Application of low-energy scanning transmission electron microscopy for the study of Pt-nanoparticle uptake in human colon carcinoma cells.
- Blank H, Schneider R, Gerthsen D, Gehrke H, Jarolim K, Marko D.Author information Laboratorium für Elektronenmikroskopie, Karlsruher Institut für Technologie (KIT) , 76128 Karlsruhe , Germany.AbstractAbstract High-angle annular dark-field scanning transmission electron microscopy (HAADF STEM) in a scanning electron microscope facilitates the acquisition of images with high chemical sensitivity and high resolution. HAADF STEM at low electron energies is particularly suited to image nanoparticles (NPs) in thin cell sections which are not subjected to poststaining procedures as demonstrated by comparison with bright-field TEM. High membrane contrast is achieved and distinction of NPs with different chemical composition is possible at first sight. Low-energy HAADF STEM was applied to systematically study the uptake of Pt-NPs with a broad size distribution in HT29 colon carcinoma cells as a function of incubation time and incubation temperature. The cellular dose was quantified, that is, the amount and number density of NPs taken up by the cells, as well as the particle-size distribution. The results show a strong dependence of the amount of incubated NPs on the exposure time which can be understood by considering size-dependent diffusion and gravitational settling of the NPs in the cell culture medium.

- The versatile biopolymer chitosan: potential sources, evaluation of extraction methods and applications.
- Kaur S, Dhillon GS.Author information Department of Mycology & Plant Pathology, Institute of Agricultural Sciences, Banaras Hindu University (BHU) , Varanasi, Uttar Pradesh , India and.AbstractAbstract Among the biopolymers, chitin and its derivative chitosan (CTS) have been receiving increasing attention. Both are composed of randomly distributed β-(1-4)-linked d-glucosamine and N-acetyl glucosamine units. On commercial scale, CTS is mainly obtained from the crustacean shells. The chemical methods employed for extraction of CTS from crustacean shells are laden with many disadvantages. Waste fungal biomass represents a potential biological source of CTS, in fact with superior physico-chemical properties, such as high degree of deacetylation, low molecular weight, devoid of protein contamination and high bioactivity. Researchers around the globe are attempting to commercialize CTS production and extraction from fungal sources. Fungi are promising and environmentally benign source of CTS and they have the potential to completely replace crustacean-derived CTS. Waste fungal biomass resulting from various pharmaceutical and biotechnological industries is grown on inexpensive agro-industrial wastes and its by-products are a rich and inexpensive source of CTS. CTS is emerging as an important natural polymer having broad range of applications in different fields. In this context, the present review discusses the potential sources of CTS and their advantages and disadvantages. This review also deals with potential applications of CTS in different fields. Finally, the various attributes of CTS sought in different applications are discussed.

- Residue and risk assessment of pyridaben in cabbage.
- Liu C, Lu D, Wang Y, Huang J, Wan K, Wang F.Author information Public Monitoring Center for Agro-product of Guangdong Academy of Agricultural Sciences, Guangzhou 510640, China; Key Laboratory of Testing and Evaluation for Agro-product Safety and Quality, Ministry of Agriculture, China.AbstractThe dissipation and residue of pyridaben in cabbage under field conditions were investigated. A sensitive, simple, and fast method for determining pyridaben in cabbage was established by high-performance liquid chromatography tandem mass spectrometry. The average recoveries were in the range of 90.29-95.00% with relative standard deviations ranging from 1.72% to 6.39%. The field results showed that pyridaben dissipated rapidly in cabbage and had a half-life of 2.8-3.5d. During harvest, the terminal residues of pyridaben were 0.01-0.80mg/kg. Given that no maximum residue limit (MRL) has been set for pyridaben in cabbage, risk assessment was evaluated by using the risk quotient (RQ). Results indicated that the RQ value was significantly lower than RQ=1. Thus, the effect of pyridaben in cabbage at the recommended dosage was negligible to humans. This study could provide guidance for the safe and reasonable use of pyridaben as a broad-spectrum acaricide and serve as a reference for the establishment of an MRL in China.
