- Comparison of visual assessments versus planimetry assessments in a large-scale clinical trial of onychomycosis.

However, no published comparison of visual assessment with planimetry is currently available on onychomycosis to support this recommendation. Objectives: This article evaluates efficacy based on visual assessment compared to planimetry measures during the course of an 84-week large-scale, single-site trial of subungual dermatophyte onychomycosis, to determine the validity of planimetry versus visual assessment. Methods: Visually assessed percentage of affected area of the target toenail was compared with planimetry calculated measurements made from digital photographs. Results: Comparison showed good statistical agreement of visual and planimetry measures based on correlation coefficient. Visual and planimetry assessments at week 84 were within 10% of each other in 92% of cases, and within 5% of each other in 74% of comparisons. However, errors in visual assessment artificially inflated the cure rates by 9% and 11% at weeks 84 and 48, respectively. Furthermore, the significant difference in effective cure between regimens using visual methods loses significance when planimetry measures are used. Conclusions: The results suggest that objective measures such as planimetry will reduce the impact of visual assessment errors, and techniques to increase the standardization of onychomycosis assessment are warranted.

Multilinear regression analysis of the biomass productivity data showed that for SA and CD the biomass productivity was independent of the proportion of medium (MWC), while the growth of CV and CR slowed down in mixtures with high proportions of wastewater. However, the biomass productivity of SA was dependent on pH, while the growth of the other microalgae was independent of pH (7-9). When evaluating the influence of pH and proportion of medium, CD appeared most robust among the algae species, despite its lower biomass productivity. All the four species reduced 80-90% of the nitrate [Formula: see text] and 60-70% of the ammonia [Formula: see text] initially present in the wastewater:medium mixture, although the extent of the reduction was dependent on the initial [Formula: see text] ratio. Both SA and CV reduced ∼20-25% of the chemical oxygen demand (COD) contained in the wastewater. This study shows the remarkable influence of certain variables that are often ignored in the search for optimal conditions of microalgal growth and also reveals the importance of considering interactions among growth variables in potential applications at large scale, particularly in the field of bioremediation.

SDS-PAGE and amino acid profile suggested that the P2.4 and P4.0 might be classified as type I collagen (PSC-I) and type V collagen (PSC-V), respectively. These collagens appeared to be dense sheet-like film linked by random-coiled filaments under SEM. The denaturation and melting temperatures of PSC-V (35.92 and 122.86°C) were significantly higher than PSC-I (32.52 and 116.01°C) assessed by CD and DSC, which could be attributed to its higher imino acid content (23.43%) and degree of hydroxylation (52.18%). FTIR confirmed their triple helical structure, and indicated more intermolecular crosslinks in PSC-I and more hydrogen bond in PSC-V. These results provide some basis for their large-scale production and further application as alternatives to mammalian collagen.

Recently, genome-wide association studies (GWAS) have been used to investigate AD pathogenesis. These GWAS have yielded important new insights into the genetic mechanisms of AD. However, these newly identified AD susceptibility loci exert only very small risk effects and cannot fully explain the underlying AD genetic risk. We hypothesize that combining the findings from different AD GWAS may have greater power than genetic analysis alone. To identify new AD risk factors, we integrated findings from 3 previous large-scale AD GWAS (n = 14,138) using a gene-based meta-analysis and subsequently conducted a pathway analysis using the kyoto encyclopedia of genes and genomes and gene ontology databases. Interestingly, we not only confirmed previous findings, but also highlighted, for the first time, the involvement of cardiovascular disease-related pathways in AD. Our results provided the clues as to the link between these diseases using pathway analysis methods. We believe that these findings will be very useful for future genetic studies of AD.
