Troponin C is a part of the troponin complex. It contains four calcium-binding EF hands. It is a component of thin filaments (along with actin and tropomyosin). It contains an N lobe and a C lobe. The C lobe serves a structural purpose and binds to the N domain of TnI. The C lobe can bind either Ca2+ or Mg2+. The N lobe, which binds only Ca2+, is the regulatory lobe and binds to the C domain of TnI after calcium binding.
The tissue specific subtypes are:
- Slow troponin C, TNNC1 (3p21.3-p14.3, Online 'Mendelian Inheritance in Man' (OMIM) 191040)
- Fast troponin C, TNNC2 (20q12-q13.11, Online 'Mendelian Inheritance in Man' (OMIM) 191039)

- Restrictive cardiomyopathy is a rare form of pediatric cardiac disease, for which the known genes include MYH7, TNNT2, TNNI3, ACTC1, and DES. We describe a pediatric proband with fatal restrictive cardiomyopathy associated with septal hypertrophy and compound heterozygosity for TNNC1 mutations (NM_0

- Restoration of blood flow to the ischemic myocardium is imperative to avoid demise of cardiomyocytes, but is paradoxically associated with irreversible damage to cardiac tissues due to the excessive generation of reactive oxygen species (ROS). - Skeletal muscle dysfunction occurs in many diseases and can lead to muscle weakness and premature muscle fatigue. Here we show that the fast skeletal troponin activator, CK-2066260, counteracts muscle weakness by increasing troponin Ca2+ affinity, thereby increasing myofibrillar Ca2+ sensitivity. Ex
