Abrin, a natural biotoxin that is homologous to ricin yet more lethal, has high potential for becoming a weapon of bioterrorism given its ease of production. Using an immobilization strategy that implements non-natural amino acids for site-specific conjugation, we have created a reusable N-methyltryptophan oxidase based magnetic bead system that is capable of detecting L-abrine, a marker for abrin poisoning, at concentrations as low as 4μM in mock urine. Furthermore, we propose that this detection strategy may be readily adaptable for sensing other targets of interest. This unique diagnostic test for abrin poisoning has demonstrated key benefits of portability and simple visual readout. These significant advantages can thus provide the potential for more rapid assessment and corresponding poison management if dedicated toxicology laboratories are not an option.

Herein, we develop a simple and highly sensitive chemiluminescence (CL) biosensing platform for site-specific determination of DNA methylation using Exonuclease III (Exo III)-assisted target recycling signal amplification. After bisulfite treatment of mixture of methylated DNA and unmethylated DNA, methylated DNA can hybridize with fluorescein (FAM)-labeled probe DNA to form double-stranded DNA (dsDNA), removing the FAM-labeled probe DNA from the surface of grapheme oxide, and the chemiluminescence resonance energy transfer (CRET) sensing signal can be observed and then amplified using Exo III-based recycling strategy. The biosensing platform exhibits excellent high sensitivity, and it can ever distinguish as low as 0.002% methylation level from the mixture, which is superior to most currently reported methods used for DNA methylation assay. In addition, the proposed method can also be used to sensitively assay MTase activity with determination limit of 0.007U/mL. This CL biosensing offers the advantages of being facile, sensitive, rapid and cost-effective. These features make the system promising for future use for early cancer diagnosis and discover of new anticancer drugs.

Using it as an electrochemical indicator, a signal-on and label-free electrochemical biosensor was developed for picomolar determination of nuclear factor kappa B (NF-κB) in serum. Initially, linear double-stranded DNA (dsDNA) probes, which contain a protein-binding site specific to NF-κB, were self-assembled on the surface of a glass carbon electrode (GCE). If the NF-κB was absent, the dsDNA probes were cut into ss-DNA fragments by the digestion of ExoIII, resulting in a low electrochemical signal of MNA due to the weak binding affinity of MNA to ss-DNA. On the contrary, in the presence of NF-κB, it could bind with the dsDNA probes at the specific site and hinder the digestion of ExoIII, resulting in a significant increase of electrochemical response due to the intercalation of MNA into the dsDNA probes. By employing the above strategy, this sensor could detect as low as 40 pM NF-κB with high specificity. To the best of our knowledge, the proposed sensor is the first attempt to use acridone derivative as an electrochemical indicator for NF-κB detection, which may represent a promising path toward clinical diagnosis and drug developments.
