A novel method that relies on the principle of acoustic sound analysis was developed to enable examination of the longitudinal progress of osseointegration. The method is based on a magnetic sphere inside a hollow cylinder of the implant. By excitation using an external magnetic field, collision of the sphere inside the implant produces a sound signal. Custom-made titanium implants equipped thusly were inserted in each lateral femoral epicondyle of 20 New Zealand White Rabbits. Two groups were investigated: Uncoated, machined surface versus antiadhesive surface; and calcium phosphate-coated surface versus antiadhesive surface. The sound analysis was performed postoperatively and weekly. After 4 weeks, the animals were euthanized, and the axial pull-out strengths of the implants were determined. A significant increase in the central frequency was observed for the loose implants (mean pull-out strength 21.1 ± 16.9 N), up to 6.4 kHz over 4 weeks. In comparison, the central frequency of the osseointegrated implants (105.2 ± 25.3 N) dropped to its initial value. The presented method shows potential for monitoring the osseointegration of different implant surfaces and could considerably reduce the number of animals needed for experiments. © 2014 Orthopaedic Research Society. The images, waveforms of pulse voltage and discharge current, and the optical emission spectra of the discharges are recorded. The rotational and vibrational temperatures of plasma are determined by comparing the simulated spectra with the experimental spectra. The effects of pulse peak voltage, pulse repetition rate and quartz tube diameter on the emission intensities of N2 (C(3)Πu→B(3)Πg, 0-0) and N2(+)B(2)Σu(+)→X(2)Σg(+),0-0 and the rotational and vibrational temperatures have been investigated. It is found that the uniform plasma with low gas temperature can be obtained, and the emission intensities of N2 (C(3)Πu→B(3)Πg, 0-0) and N2(+)B(2)Σu(+)→X(2)Σg(+),0-0 rise with increasing the pulse peak voltage and pulse repetition rate, while decrease as the increase of quartz tube diameter. In addition, under the condition of 28kV pulse peak voltage, 150Hz pulse repetition rate and 7mm quartz tube diameter, the plasma gas temperature is determined to be 330K. The results also indicate that the plasma gas temperature keep almost constant when increasing the pulse peak voltage and pulse repetition rate but increase with the increase of the quartz tube diameter.

Wistar rats were injected unilaterally with lactacystin (2.5μg/2μl) or 6-OHDA (8μg/2μl) into the substantia nigra (SN) pars compacta. Four weeks after the lesion, the animals were treated chronically with l-DOPA (25 or 50mg/kg) for two weeks. During l-DOPA treatment, the lactacystin-treated rats were tested for catalepsy and forelimb asymmetry. Rotational behavior was evaluated after apomorphine (0.25mg/kg) and l-DOPA in both PD models. After completion of experiments, the animals were killed and the levels of DA and its metabolites in the striatum and SN were assayed. We found that acute l-DOPA administration effectively decreased catalepsy and increased the use of the compromised forelimb in the cylinder test. However, the lactacystin group did not respond to apomorphine or acute l-DOPA administration in the rotational test. Repeated l-DOPA treatment produced contralateral rotations in both PD models, but the number of rotations was much greater in the 6-OHDA-lesioned rats. Both toxins markedly (>90%) reduced the levels of DA and its metabolites in the striatum and SN, while l-DOPA diminished these decreases, especially in the SN. By demonstrating the efficacy of l-DOPA in several behavioral tests, our study confirms the usefulness of the lactacystin lesion as a model of PD. However, marked differences in the rotational response to apomorphine and l-DOPA suggest different mechanisms of neurodegeneration evoked by lactacystin and 6-OHDA.

The purpose of this study was to examine the effect of TS on healthy and hemiparkinsonian adult rats. Therefore, the animals received TS for 14 days and 15min three times daily. At different time points rats were tested in various behavioral tests (amphetamine-induced rotation, cylinder test, staircase test). Finally, rats were sacrificed, their brains removed, and processed for Golgi-Cox analyses, tyrosine hydroxylase immunohistochemistry and quantitative RT-PCR. We found that the striatal 6-OHDA lesion itself induced a long-term increase of astroglial Fgf2 transcript levels, but was not further increased by TS. In contrast TS applied to healthy rats elicited a transient short-term increase of Fgf2 in the striatum and Bdnf, Grin1, and Fgf2 in the hippocampus. Moreover, behavioral and histological analyses do not support a beneficial effect of TS for hemiparkinsonian rats, applied for two weeks starting one day after partial striatal 6-OHDA lesion.
