- General holistic impairment in congenital prosopagnosia: Evidence from Garner's speeded-classification task.
- Tanzer M1, Freud E, Ganel T, Avidan G.Author information 1a Department of Psychology , Ben-Gurion University of the Negev , Beer Sheva , Israel.AbstractCongenital prosopagnosia (CP), a lifelong impairment in face processing in the absence of brain damage, is often ascribed to impairment in holistic processing. It is still debated whether such difficulties are restricted to faces or whether they can also be observed for nonfacial stimuli. Here, we investigate this issue by examining CP individuals and their controls on two variations of the Garner speeded classification task tailored to assess holistic processing of nonfacial stimuli. In Experiment 1, participants were asked to judge the width of visually presented rectangles while ignoring their irrelevant height, or to judge changes in width while height remained constant. Critically, while controls exhibited the expected Garner interference, no such interference was observed for the CPs, indicating impaired holistic processing of integral, nonfacial shape dimensions. Experiment 2, utilized the same Garner paradigm, but here participants were asked to judge integral dimensions that are unrelated to shape (colour). Importantly, both CPs and controls exhibited the same level of Garner interference, indicating intact integral processing of colour dimensions. This dissociation between the performance on the two Garner tasks indicates that CPs do not exhibit a general local processing bias or impaired integration of any perceptual dimensions, but rather a deficit that is restricted to tasks requiring holistic integral perception of shape dimensions. Taken together, these results provide evidence for the existence of a general impairment in holistic shape perception in CP, which may be related to the mechanisms underlying this disorder.

In this work, a novel plasmonic based optical biosensor prototype for ochratoxin A is described. It exploits the metal-enhanced fluorescence phenomenon due to the silver film over nanosphere plasmonic substrate. Since ochratoxin A could be present in different food commodities, sensor performances have been tested on three different matrices (dried milk, juices, and wheat mix). Firstly, a common OTA extraction solvent and a labeling and detection protocol were defined for the analyzed matrices. Then, the efficiency of the Ag-FON surfaces in signal amplification for the detection of low ochratoxin A concentrations was defined. Using samples spiked with OTA-AF 647 or with unlabeled OTA we were able to detect the mycotoxin at concentrations lower than E.U. specifications of 0.5μg/kg in wheat, milk and apple juice. The test performances are comparable to those of ELISA kits but the platform presented here, once optimized, present some perspective advantages, such as: low cost and time consuming, versatility of the protocol for the investigation of different matrices, employment also in non-qualified laboratories, small dimensions that allow its integration in a compact device for OTA on-site detection.

Electronic address: agil@ua.pt.AbstractThis paper describes a proton high resolution magic angle spinning (HRMAS) nuclear magnetic resonance (NMR) metabolomic study of lettuce (Lactuca sativa L.) leaves to characterise metabolic adaptations during leaf growth and exposure to mancozeb. Metabolite variations were identified through multivariate analysis and checked through spectral integration. Lettuce growth was accompanied by activation of energetic metabolism, preferential glucose use and changes in amino acids, phospholipids, ascorbate, nucleotides and nicotinate/nicotinamide. Phenylalanine and polyphenolic variations suggested higher oxidative stress at later growth stages. Exposure to mancozeb induced changes in amino acids, fumarate and malate, suggesting Krebs cycle up-regulation. In tandem disturbances in sugar, phospholipid, nucleotide and nicotinate/nicotinamide metabolism were noted. Additional changes in phenylalanine, dehydroascorbate, tartrate and formate were consistent with a higher demand for anti-oxidant defence mechanisms. Overall, lettuce exposure to mancozeb was shown to have a significant impact on plant metabolism, with mature leaves tending to be more extensively affected than younger leaves.

These adverse conditions frequently affect HIV and tuberculosis patients on regular medication in resource-poor settings. Here, we report full integration of sample preparation with the read-out of a 5-parameter liver assay panel (LAP) on a portable, easy-to-use, fast and cost-efficient centrifugal microfluidic analysis system (CMAS). Our unique, dissolvable-film based centrifugo-pneumatic valving was employed to provide sample-to-answer fashion automation for plasma extraction (from finger-prick of blood), metering and aliquoting into separate reaction chambers for parallelized colorimetric quantification during rotation. The entire LAP completes in less than 20min while using only a tenth the reagent volumes when compared with standard hospital laboratory tests. Accuracy of in-situ liver function screening was validated by 96 separate tests with an average coefficient of variance (CV) of 7.9% compared to benchtop and hospital lab tests. Unpaired two sample statistical t-tests were used to compare the means of CMAS and benchtop reader, on one hand; and CMAS and hospital tests on the other. The results demonstrate no statistical difference between the respective means with 94% and 92% certainty of equivalence, respectively. The portable platform thus saves significant time, labour and costs compared to established technologies, and therefore complies with typical restrictions on lab infrastructure, maintenance, operator skill and costs prevalent in many field clinics of the developing world. It has been successfully deployed to a centralised lab in Nigeria.
