Here, we present evidence that microglia control Listeria infection differently than macrophages. Infection of primary microglial cultures and murine cell lines with Listeria resulted in a dual function of the two gene expression programmes involved in early and late immune responses in macrophages. Whereas the bacterial gene hly seems responsible for both transcriptional programmes in macrophages, Listeria induces in microglia only the tumor necrosis factor (TNF)-regulated transcriptional programme. Listeria also represses in microglia the late immune response gathered in two clusters, microbial degradation, and interferon (IFN)-inducible genes. The bacterial gene actA was required in microglia to induce TNF-regulated responses and to repress the late response. Isolation of microglial phagosomes revealed a phagosomal environment unable to destroy Listeria. Microglial phagosomes were also defective in several signaling and trafficking components reported as relevant for Listeria innate immune responses. This transcriptional strategy in microglia induced high levels of TNF-α and monocyte chemotactic protein-1 and low production of other neurotoxic compounds such as nitric oxide, hydrogen peroxide, and Type I IFNs. These cytokines and toxic microglial products are also released by primary microglia, and this cytokine and chemokine cocktail display a low potential to trigger neuronal apoptosis. This overall bacterial strategy strongly suggests that microglia limit Listeria inflammation pattern exclusively through TNF-mediated responses to preserve brain integrity. GLIA 2014;62:233-246.

In this study, the roles of extracellular chitinase and an antifungal compound produced by strain C61 were investigated to elucidate their contributions to biological control activity. The bacterium possessed a locus chi54 encoding an extracellular chitinase, and mutation of chi54 eliminated chitinase production. Production of the extracellular enzyme and expression of the chi54 transcript were increased in the wild-type strain when chitin was added to the culture medium. In vitro assays showed that purified chitinase inhibited spore germination of multiple pathogens. However, the in planta biocontrol activity of filtrates of cultures grown in the presence of chitin was lower than that of filtrates grown without chitin, indicating that correlation between chitinase and biocontrol activity was lacking. The analysis of C61 culture filtrates revealed an antifungal cyclic lipopeptide, chromobactomycin, whose structure contained a unique nonameric peptide ring. The purified chromobactomycin inhibited the growth of several phytopathogenic fungi in vitro, and plant application significantly reduced disease severity for several pathogens. Furthermore, the production of chromobactomycin was reduced in cultures amended with chitin. These data suggest that the production of both the extracellular chitinase Chi54 and the newly identified antibiotic chromobactomycin can contribute, in an interconnected way, to the suppression of plant disease by Chromobacterium sp. strain C61.

We show here that expression of MT1a, MT2b1, MT2b2 and MT3 in barley leaves increased more than 50-fold during the first 10 d after germination. Concurrently, the root-specific gene MT1b1 was 1000-fold up-regulated. Immunolocalizations provided the first evidence for accumulation of MT1a and MT2a proteins in planta, with correlation to transcript levels. In developing grains, MT2a and MT4 expression increased 4- and 300-fold over a 28-day-period after pollination. However, among the MT grain transcripts MT2c was the most abundant, whereas MT4 was the least abundant. Excess Cu up-regulated three out of the six MTs expressed in leaves of young barley plants. In contrast, most MTs were down-regulated by excess Zn or Cd. Zn starvation led to up-regulation of MT1a, whereas Cu starvation up-regulated MT2a, which has two copper-responsive elements in the promoter. Arabidopsis lines constitutively overexpressing barley MT2a showed increased sensitivity to excess Cd and Zn but no Cu-induced response. We suggest that barley MTs are differentially involved in intracellular homeostasis of essential metal ions and that a subset of barley MTs is specifically involved in Cu detoxification.
