The dual-coated SR pellets composed of a drug-loaded nonpareil core, a sub-coating layer (HPMC 6cps) and an SR-coating layer (Aquacoat® ECD, Eudragit® RS 30D or Kollicoat® SR 30D) were prepared by a bottom-spray fluidized bed-coating method. The drug release from the dual-coated SR pellets coated with Aquacoat® ECD followed a zero-order profile in water, and the drug release was not affected by the coating level of the sub-coating layer and stable under the accelerated storage condition (40 °C, 75% RH) for 6 months. The CR preparation showed significantly decreased values of maximum drug concentration (Cmax) and elimination rate (K) than the reference product (LEVOTUS® SYR) but the similar bioavailability (F = 95.43%). The novel CR preparation presents promising delivery of LDP with an immediate and sustained release manner, with similar clinical effect as the commercial IR product.

The analytical method was validated by detection limits, precision, accuracy and recovery experiments, obtaining satisfactory results in all cases. Results of both minor and trace elements were found mostly similar in all varieties of milk and yogurts studied. Copper and manganese were comparatively high in fruit mixed yogurts, while selenium was high in milk samples. The overall concentrations of minor elements were found to have good nutritional values in accordance to RDA. Zn showed the highest concentration, followed by Rb, Se, Sr and Cr. The level of toxic trace elements, including As, Cd and Pb were very low and did not pose any threat to consumers.

Electronic address: anna.szymczycha@pwr.wroc.pl.AbstractSix sample preparation procedures, i.e., the total decomposition in a HNO3 and H2O2 mixture by microwave-assisted or hot-plate heating, a partial decomposition by means of solubilisation in aqua regia or tetramethyl ammonium hydroxide and simple dissolution in diluted HNO3 or in H2O, for the determination of the total content of Ba, Ca, Cd, Cr, Cu, Fe, Mg, Mn, Ni, P, Pb, Sr and Zn in slim instant coffees using inductively coupled plasma optical emission spectrometry were compared. The performance of procedures was determined based on the precision and the accuracy of results and limits of detection of elements. It was found that the extraction with aqua regia provides the best results, i.e., limits of detection of elements within 0.11-108ngmL(-1), the precision of 0.6-5% and the accuracy better than 5%. Elements' concentrations agree with those after the wet digestion in the HNO3 and H2O2 mixture, therefore this procedure is a simple and fast alternative to complete sample decomposition procedures and can be easily applied for routine analyses. Six commercial slim coffee products were analysed with the proposed procedure.
