• Pena-Shokeir syndromeとMarden-Walker syndromeは関節拘縮と先天性の多臓器奇形を伴う進行性の運動ニューロン変性疾患である。(NELSON)

• 遺伝性があることが知られており遺伝形式は常染色体劣性遺伝である。
• 症例のうち50%程度に遺伝性が認められ、全例で遺伝性が見られるわけではない。
• I型では多発性関節拘縮、顔面異常、および肺低形成が特徴的であり、遺伝形質は常染色体劣性遺伝、有病率は100万出生に1未満である。胎児の約30％が子宮内で死亡し、新生児の大多数は肺低形成のために新生児期初期に死亡しているとされる(https://www.ncbi.nlm.nih.gov/pmc/articles/PMC6207248/)。
• II型は脳・眼・顔・骨格症候群(COFS症候群)とも呼ばれ、脳内石灰化、白内障、微小角膜、視神経萎縮、進行性関節拘縮、および成長不全が特徴的で、進行性の脳萎縮を伴う。常染色体劣性遺伝で、5歳までに死に至ることが多い(https://www.ncbi.nlm.nih.gov/pmc/articles/PMC6207248/)
