The biosensor consists of straight gold waveguides embedded in CYTOP with an etched fluidic channel. The gold waveguides were functionalized with immunoglobulin G against blood group A (anti-A IgG) by forming a self-assembled monolayer (SAM) of 16-mercaptohexadecanoic acid (16-MHA) and then conjugating the anti-A IgG through carbodiimide chemistry. In order to demonstrate anti-A surface selectivity, solutions of O-type, B-type, A-type and AB-type red blood cells (RBCs) were sequentially injected over an anti-A functionalized waveguide. Surfaces were regenerated by lysing attached cells with distilled/deionized water (DDI H2O). The efficiency of surface regeneration with DDI H2O was very high as determined by performing six sequential binding/regeneration cycles of A RBC capture on the same anti-A surface. Also, five solutions of different A RBC concentrations, ranging from 1.14×10(5)cells/ml to 1.83×10(6)cells/ml, were injected over an anti-A surface to determine the limit of detection (LOD), which was found to be less than 3×10(5)cells/ml. Finally, the response produced by a single cell bound to a waveguide was determined by relating the number of bound cells to the response produced, from which the signal-to-noise ratio for single cell detection was determined to be ~95. The waveguides are promising as simple, low-cost and compact transducers, functionalized using standard thiol-based chemistries, for the selective detection of cells.

Morphological changes of the PPy and CNT modified electrodes were investigated using scanning electron microscopy. The electrochemical behavior of the bienzymatic electrode (NaR-SOD1-CNT-PPy-Pt) was characterized by cyclic voltammetry exhibiting quasi-reversible redox peak at +0.06 V and reversible redox peaks at -0.76 and -0.62V vs. Ag/AgCl, for the immobilized SOD1 and NaR respectively. The electrocatalytic activity of SOD1 towards NO2(-) oxidation observed at +0.8 V was linear from 100 nM to 1mM with a detection limit of 50 nM and sensitivity of 98.5 ± 1.7 nA µM(-1)cm(-2). Similarly, the coimmobilized NaR showed its electrocatalytic activity towards NO3(-) reduction at -0.76 V exhibiting linear response from 500 nM to 10mM NO3(-) with a detection limit of 200 nM and sensitivity of 84.5 ± 1.56 nA µM(-1)cm(-2). Further, the present bienzymatic biosensor coated with cellulose acetate membrane for the removal of non-specific proteins was used for the sensitive and selective determinations of NO2(-) and NO3(-) present in human plasma, whole blood and saliva samples.

Various lymphoid tissues from immunized donors, such as lymph nodes or peripheral blood lymphocytes from individuals with tumor or lymphocytes infiltrating tumor masses may serve as a source of specific anti-tumor antibody repertoire for generation of tumor-focused phage display libraries. In the case of lack of tumor-associated antigens in the purified form, high affinity anti-tumor antibodies can be isolated through library panning on whole cells expressing these antigens. However, affinity selection against cell surface specific antigens within highly heterogeneous population of molecules is not a very efficient process that often results in the selection of unspecific antibodies or antibodies against intracellular antigens that are generally useless for targeted immunotherapy. In this work, we developed a new cell-based antibody selection protocol that, by eliminating the contamination of dead cells from the cell suspension, dramatically improves the selection frequency of anti-tumor antibodies recognizing cell surface antigens.
