These are controlled by a network architecture that comprises two regulatory factors: posttranslational oscillation (PTO) and a transcription/translation feedback loop (TTFL). The clock proteins KaiA, KaiB, and KaiC are essential for the circadian rhythm of the unicellular species Synechococcus elongatus PCC 7942. Temperature-compensated autonomous cycling of KaiC phosphorylation has been proposed as the primary oscillator mechanism that maintains the circadian clock, even in the dark, and it controls genome-wide gene expression rhythms under continuous-light conditions (LL). However, the kaiC(EE) mutation (where "EE" represents the amino acid changes Ser431Glu and Thr432Glu), where phosphorylation cycling does not occur in vivo, has a damped but clear kaiBC expression rhythm with a long period. This suggests that there must be coupling between the robust PTO and the "slave" unstable TTFL. Here, we found that the kaiC(EE) mutant strain in LL was hypersensitive to the dark acclimation required for phase shifting. Twenty-three percent of the genes in the kaiC(EE) mutant strain exhibited genome-wide transcriptional rhythms with a period of 48 h in LL. The circadian phase distribution was also conserved significantly in most of the wild-type and kaiC(EE) mutant strain cycling genes, which suggests that the output mechanism was not damaged severely even in the absence of KaiC phosphorylation cycles. These results strongly suggest that the KaiC phosphorylation cycle is not essential for generating the genome-wide rhythm under light conditions, whereas it is important for appropriate circadian timing in the light and dark.

- Warren-Smith SC, Monro TM.AbstractBragg gratings have been written in exposed-core microstructured optical fibers for the first time using a femtosecond laser. Second and third order gratings have been written and both show strong reflectivity at 1550 nm, with bandwidths as narrow as 60 pm. Due to the penetration of the guided field outside the fiber the Bragg reflections are sensitive to the external refractive index. As different modes have different sensitivities to refractive index but the same temperature sensitivity the sensor can provide temperature-compensated refractive index measurements. Since these Bragg gratings have been formed by physical ablation, these devices can also be used for high temperature sensing, demonstrated here up to 800°C. The fibers have been spliced to single mode fiber for improved handling and integration with commercial interrogation units.

- Thermal sensitivity of DASH interferometers: the role of thermal effects during the calibration of an Echelle DASH interferometer.
- Marr KD, Englert CR, Harlander JM, Miller KW.AbstractThe use of a Doppler asymmetric spatial heterodyne (DASH) interferometer with an Echelle grating provides the ability to simultaneously image the 558 and 630 nm emission lines (e.g., at grating orders of n=8 and n=7, respectively) of atomic oxygen in the thermosphere. By measuring the Doppler shifts of these lines (expected relative change in wavelength on the order of 10⁻⁸), we are able to determine the thermospheric winds. Because the expected wavelength changes due to the Doppler shift are so small, understanding, monitoring, and accounting for thermal effects is expected to be important. Previously, the thermal behavior of a temperature-compensated monolithic DASH interferometer was found to have a higher thermal sensitivity than predicted by a simple model [Opt. Express 18, 26430, 2010]. A follow-up study [Opt. Express 20, 9535, 2012] suggested that this is due to thermal distortion of the interferometer, which consists of materials with different coefficients of thermal expansion. In this work, we characterize the thermal drift of a nonmonolithic Echelle DASH interferometer and discuss the implications of these results on the use of only a single wavelength source during calibration. Furthermore, we perform a finite element analysis of the earlier monolithic interferometer in order to determine how distortion would affect the thermal sensitivity of that device. Incorporating that data into the model, we find good agreement between the modified model and the measured thermal sensitivities. These findings emphasize the fact that distortion needs to be considered for the design of thermally compensated, monolithic DASH interferometers.
