However, because of high first-pass metabolism butorphanol is not available in market as oral dosage form. Drugs that undergo extensive first-pass metabolism can be delivered orally if protected in the stomach, and proximal small intestine. An oral controlled porosity osmotic pump (CPOP) was designed to deliver butorphanol tartrate that can maintain therapeutic blood concentration up to 24 h. The target release profile for extended release formulation was calculated by Wagner Nelson de-convolution using published immediate release blood concentration data for oral route. Composition of the core and coating were optimized using USFDA approved ingredients by evaluation of the drug release. Drug release from the developed system was inversely proportional to the weight gain and directly related to the level of pore former. Scanning electron microscopy (SEM) confirmed the formation of pores in the coating membrane on contact with water which lead to drug to release. Kinetic models were applied to drug release data to establish the drug release mechanism. The developed osmotic system effectively delivers selected drug at a predetermined rate for extended period.

The aim of this study was to design a controlled porosity osmotic pump (CPOP) of diltiazem hydrochloride to deliver the drug in a controlled manner. CPOP tablets were prepared by incorporation of drug in the core and subsequent coating with cellulose acetate as semi-permeable membrane. Non-ionic surfactants were applied as pore-formers as well. The effect of pore-formers concentration on the in vitro release of diltiazem was also studied. The formulations were compared based on four comparative parameters, namely, total drug released after 24 h (D24 h), lag-time (tL), squared correlation coefficient of zero order equation (RSQzero) and mean percent deviation from zero order kinetic (MPDzero). Results of scanning electron microscopy studies exhibited formation of pores in the membrane from where the drug release occurred. It was revealed that drug release rate was directly proportional to the concentration of the pore-formers. The value of D24 h in the formulations containing Tween 80 (10%) and Brij 35 (5%) were found to be more than 94.9%, and drug release followed zero order kinetic (RSQzero > 0.99 and MPDzero < 8%) with acceptable tL (lower than 1 h).

A series of OMCN-x samples with different pyrolysis temperatures have been reported. The formation of these composite materials was verified by detailed characterization (e.g., Fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy, N2 adsorption, X-ray diffraction, scanning electron microscopy, and transmission electron microscopy). The results showed that the materials were structurally well ordered with two-dimensional porous structure, high surface area and large pore volume. The influence of BET surface area and different amounts of N-bonding configurations formed at different pyrolysis temperatures of OMCN-x for the electrocatalysis towards hydrogen peroxide, nitrobenzene, and nicotinamide adenine dinucleotide were investigated in detail. Results indicated that OMCN treated at 800°C with largest BET surface area and highest amounts of pyrindinic N showed improved electrocatalytic activity for H2O2, nitrobenzene, and NADH in neutral solution.
