Electronic address: carmen.horrillo.guemes@csic.es.AbstractThe artificial olfaction, based on electronic systems (electronic noses), includes three basic functions that operate on an odorant: a sample handler, an array of gas sensors, and a signal-processing method. The response of these artificial systems can be the identity of the odorant, an estimate concentration of the odorant, or characteristic properties of the odour as might be perceived by a human. These electronic noses are bio inspired instruments that mimic the sense of smell. The complexity of most odorants makes characterisation difficult with conventional analysis techniques, such as gas chromatography. Sensory analysis by a panel of experts is a costly process since it requires trained people who can work for only relatively short periods of time. The electronic noses are easy to build, provide short analysis times, in real time and on-line, and show high sensitivity and selectivity to the tested odorants. These systems are non-destructive techniques used to characterise odorants in diverse applications linked with the quality of life such as: control of foods, environmental quality, citizen security or clinical diagnostics. However, there is much research still to be done especially with regard to new materials and sensors technology, data processing, interpretation and validation of results. This work examines the main features of modern electronic noses and their most important applications in the environmental, and security fields. The above mentioned main components of an electronic nose (sample handling system, more advanced materials and methods for sensing, and data processing system) are described. Finally, some interesting remarks concerning the strengths and weaknesses of electronic noses in the different applications are also mentioned.

Unexpectedly, the fusion proteins self-assembled into insoluble active particles as the ELPs underwent a hardly reversible phase transition. The specific activity of the particles was 92% of the native counterparts, which means it can act as a pull-down handler for converting soluble proteins into active aggregates. We evaluated the characterizations of the insoluble active xylanase particles in detail and the results were encouraging. The pH optimum (6.0) of the particles was the same as the free one, but the optimum pH range was 5-7, while the free xylanase was 6-7. The free xylanase had an optimum temperature of 50°C, whereas the insoluble active xylanase particles shifted to 70°C. The pH stability, thermostability and storage stability of the xylanase particles increased significantly when compared with the free xylanase. We also observed an increase of the Km values of the free xylanase from 0.374gL(-1) to 0.980gL(-1) at the insoluble state. The considerable higher activity and stability of the xylanase particles were much like immobilized xylanases and could be valuable for its industrial application.

Convincing data exist defining transcriptionally distinct posterior fossa (PF) sub-groups A and B at diagnosis. The clinical and biological consequence of these sub-groups at recurrence has not yet been defined. Genome and transcriptome microarray profiles and clinical variables of matched primary and first recurrent PF EPN pairs were used to identify biologically distinct patterns of progression between EPN sub-groups at recurrence. Key findings were validated by histology and immune function assays. Transcriptomic profiles were partially conserved at recurrence. However, 4 of 14 paired samples changed sub-groups at recurrence, and significant sub-group-specific transcriptomic changes between primary and recurrent tumors were identified, which were predominantly immune-related. Further examination revealed that Group A primary tumors harbor an immune gene signature and cellular functionality consistent with an immunosuppressive phenotype associated with tissue remodeling and wound healing. Conversely, Group B tumors develop an adaptive, antigen-specific immune response signature and increased T-cell infiltration at recurrence. Clinical distinctions between sub-groups become more apparent after first recurrence. Group A tumors were more often sub-totally resected and had a significantly shorter time to subsequent progression and worse overall survival. Minimal tumor-specific genomic changes were observed for either PF Groups A or B at recurrence. Molecular sub-groups of PF EPN convey distinct immunobiologic signatures at diagnosis and recurrence, providing potential biologic rationale to their disparate clinical outcomes. Immunotherapeutic approaches may be warranted, particularly in Group A PF EPN.
