Here, we have investigated the expression of the four ErbB4 isoforms and we found that all of them are expressed by neural progenitor cells migrating from the subventricular zone toward the olfactory bulb through the rostral migratory stream. We quantified the absolute expression of the different ErbB4 isoforms and found that all of them are highly expressed in the regions characterized by high interneuron migration, whereas in the olfactory bulb regions, where migration stops, ErbB4 isoforms containing exon JMb and lacking exon cyt1 (called 'cyt2 isoforms') are expressed more than isoforms containing exons JMa and cyt1. Indeed, we have shown previously that neural progenitor cells stably expressing ErbB4-JMb-cyt2 have a very low migratory activity. To investigate whether the different ErbB4 isoforms confer a distinct adhesion preference for transmembrane neuregulin 1, neural progenitor cells expressing these were tested in vitro in the stripe choice assay. We found that each of the four ErbB4 isoforms is able to confer cells with an adhesion preference for cells expressing the transmembrane neuregulin 1 type III.

The Bergman minimal model provides a non-invasive approach for estimating insulin sensitivity from the glucose insulin time series data of intravenous glucose tolerance test (IVGTT). However, quite often in the traditional gradient based techniques for deriving insulin sensitivity from the minimal model, improper initialization leads to convergence problems and results in final solution, which are either incorrect or physiologically not feasible. This paper deals with a differential evolution-based approach for the determination of insulin sensitivity from the minimal model using clinical test data. Being a direct search based technique, the proposed approach enables the determination of the global solution irrespective of the initial parameter values. The fitting performance of the model with parameters estimated from the proposed approach is found to be higher than the corresponding model estimated from the widely used gradient based approach. A high correlation coefficient of 0.964 (95% confidence interval of [0.897,0.988]) is acheived between the estimated insulin sensitivity and the one obtained from the population based approach for 16 subjects. The high correlation signifies the relative similarity between the two estimated indices in representing the same physiological phenomena. The simulation results and statistical analysis reveal that the proposed technique provides a reliable estimate of insulin sensitivity with minimum prior knowledge.

The batch kinetic data were statistically analyzed with a three-layered feed-forward artificial neural network (ANN) model for predicting the phenol removal efficiency from the water environment. The input parameters considered were the adsorbent dose, initial phenol concentration, contact time, and percentage of clay and silt content in soils. The response output of the ANN model was considered as the phenol removal efficiency. The predicted results of phenol removal efficiency were compared with the experimental values as obtained from batch tests and also tests for goodness of fitting in ANN model with experimental results. The estimated values of coefficient of correlation (R = 0.99) and mean squared error (MSE = 0.006) reveals a reasonable closeness of experimental and predicted values. Out of five different types of soil, type E exhibited the highest removal efficiency (31.6 %) corresponding to 20 mg/L of initial phenol concentration. A sensitivity analysis was also carried out on the ANN model to ascertain the degree of effectiveness of various input variables.

In particular, the study determined the effect of PMm on cellular viability, 2-NBDG glucose uptake, insulin secretion, and NF-κB translocation in mouse pancreatic insulinoma cells (NIT-1). Furthermore, in vivo acute toxicity and antidiabetic studies were performed using streptozotocin (STZ)-induced type 1 and STZ-nicotinamide-induced type 2 diabetic rat models to evaluate various biochemical parameters and markers of oxidative stress and pro-inflammatory cytokines. Five isoquinoline alkaloids and three phenolic compounds were tentatively identified in the PMm by LC/MS Triple TOF. The study results showed that PMm is non-toxic to NIT-1 cells and significantly increased the glucose uptake and insulin secretion without affecting the translocation of NF-κB. Moreover, the non-toxic effects of PMm were confirmed through an in vivo acute toxicity study, which revealed that the serum insulin and C-peptide levels were significantly upregulated in type 2 diabetic rats and that no significant changes were observed in type 1 diabetic rats. Similarly, PMm was found to downregulate the levels of oxidative stress and pro-inflammatory cytokines in type 2 diabetic rats by alleviating hyperglycemia. Therefore, we conclude that PMm may be developed as an antidiabetic agent for the treatment of type 2 diabetes-associated conditions.
