Electronic address: leeil@egr.msu.edu.AbstractHydrophilic nisin-loaded hydrophobic poly (lactic acid) (PLA) particles with controlled size and shape were successfully produced utilizing a one-step single emulsification method. Preliminary shear stress and temperature tests showed that there was no significant loss in the nisin inhibition activity during this process. PLA/nisin composite particles were prepared into solid nanocomposite spheres (50-200nm) or hollow microcomposite spheres (1-5μm) under the operative conditions developed in our previous study, in which the hydrophilic nisin in the aqueous phase solution could be entrapped in the hydrophobic polymer in the emulsification process generating either single or multiple emulsions. The incorporation of nisin in PLA had little effect on key processing conditions, which allows the dynamic control of the morphology and property of resulting particles. Microscopic and surface analyses suggested that nisin was dispersed uniformly inside the polymer matrix and adsorbed on the particle surface. The encapsulation amount and efficiency were enhanced with the increase in nisin loading in the aqueous solution. Unique reversible control of particle size and shape by this process was successfully applied in the nisin encapsulation. Alternating temperature in the repeating emulsification steps improved the encapsulation efficiency while generated particles in desired size and shape.

SirexAA-E CAZymes most frequently contain a carbohydrate-binding module from family 2a (CBM2a). The DNA encoding the CBM2a from gene locus SACTE_0237, the most abundantly expressed cellulase from SirexAA-E, was cloned into an Escherichia coli expression vector and expressed as a C-terminal fusion protein to GFP. The GFP-CBM2a fusion protein was purified from insoluble inclusion bodies and refolded. The solubilized protein was separated by size-exclusion chromatography into high molecular weight GFP-CBM2a multimers and monomeric GFP-CBM2a. Only the monomeric CBM2a protein was found to have high relative affinity (partition coefficient of 0.62±0.04L/g) to cellulose. Binding of monomeric CBM2a prepared in this manner exhibits fully reversible, high affinity binding to cellulose.

The present study tested the hypothesis that fetal hypoxia results in heightened brain vulnerability to hypoxic-ischemic (HI) injury in neonatal rats via down-regulation of glucocorticoid receptor (GR) in the developing brain. Time-dated pregnant rats were exposed to hypoxia (10.5% O2) from days 15 to 21 of gestation. Brain HI injury was determined in day 10 pups. Maternal hypoxia resulted in asymmetric intrauterine growth restriction in the fetus. The brain HI injury was significantly increased in maternal hypoxia-treated pups as compared with the normoxia control in both males and females. Activation of brain GR by dexamethasone injection into the right lateral ventricle produced a concentration-dependent reduction of HI-induced brain injury in control pups. Maternal hypoxia significantly decreased GR mRNA and protein abundance in the fetal brain and neonatal hippocampus and abolished the dexamethasone-mediated neuroprotective effect in pup brains. This decreased GR expression was resulted from increased DNA methylation, decreased binding of transcription factors Egr-1 and Sp1 to GR gene exon 17 and 111 promoters, and reduced expression of GR exon 17 and 111 mRNA variants. The results demonstrate that gestational hypoxia causes epigenetic repression of GR gene expression in the developing brain resulting in the heightened brain vulnerability to HI injury in neonatal rats.
