Nonetheless, this is not an easy task and often not affordable by many laboratories as it might require expensive instrumentation and sophisticated analysis software. Here, we describe a simple protocol for performing sequential immunostainings on two different Drosophila specimens. Our strategy relies on an efficient and reproducible method for removal primary antibodies and/or fluorophore-conjugated secondary antibodies that does not affect antigene integrity. We show that alternation of multiple rounds of antibody incubation and removal on the same slide, followed by registration of the same DAPI-stained image, provides a simple framework for the sequential detection of several antigens in the same cell. Given that the sample fixation procedures used for Drosophila tissues are compatible with most specimen processing protocols, we can envisage that the multicolor immunostaining strategy presented here can be also adapted to different samples including mammalian tissues and/or cells. By polystyrene (PS) microspheres based LB template method, the silver nanotriangle array with about 100nm in height was constructed on the surface of glass slide. On the surface of Ag nanotriangles array, the immune reaction of antigens and labeled antibodies was carried out. Based on the interaction of fluorophores from antibodies with the plasmon resonance from Ag nanotriangles and the enrichment effect of this patterned array, 3.11 times enhancement of the fluorescent intensity of the target antibodies was obtained. According to the fitting curve of fluorescent intensities and logarithmic concentrations of labeled antibodies from 100pg/mL to 10μg/mL, it concludes that the limit of detection by this Ag nanotriangles array for immune complex is 100pg/mL. Due to the advantages of high sensitivity, good reproducibility, and convenient fabrication, the 2D silver nanotriangles array could be an exciting platform for bioassays in proteomics, drug discovery and diagnostics.

Instead, the most-stable contact angle obtained from mechanical stimuli of the system is indeed experimentally accessible. Monitoring the susceptibility of a sessile drop to a mechanical stimulus enables to identify the most stable drop configuration within the practical range of contact angle hysteresis. Two different stimuli may be used with sessile drops: mechanical vibration and tilting. The most stable drop against vibration should reveal the changeless contact angle but against the gravity force, it should reveal the highest resistance to slide down. After the corresponding mechanical stimulus, once the excited drop configuration is examined, the focus will be on the contact angle of the initial drop configuration. This methodology needs to map significantly the static drop configurations with different stable contact angles. The most-stable contact angle, together with the advancing and receding contact angles, completes the description of physically realizable configurations of a solid-liquid system. Since the most-stable contact angle is energetically significant, it may be used in the Wenzel, Cassie or Cassie-Baxter equations accordingly or for the surface energy evaluation.
