Lipid profiling of human plasma from peritoneal dialysis patients using an improved 2D (NP/RP) LC-QToF MS method.

This 2D LC-QToF MS system was built based on a ten-port, two-position valve as the interface, the conditions of which had been optimized and discussed in detail. As two loops were used to trap and transfer the first dimensional elute to the second dimension separately, this new interface suppressed the sample band broadening in the first dimensional column, increased the recovery and repeatability of 2D LC interface, and offered the possibility for the realization of not-stop-flow NP/RP 2D LC system. Finally, 190 endogenous lipid species out of 10 lipid classes were determined within a single run from the plasma of peritoneal patients. This method was also applied to identify the difference in lipid profile between plasma from peritoneal dialysis patients with bad volume status and peritoneal dialysis patients with good volume status. The discovery of 30 potential biomarkers would be helpful to the malnutrition, inflammation, and atherosclerosis syndrome investigation.

Nevertheless, it is often quite difficult to isolate compounds with high purity because of the complexity of TCMs in chemical composition. In this work, a two-dimensional preparation method was successfully developed for the preparation of high-purity compounds from the stem of Lonicera japonica Thunb, based on two novel polar copolymerized RP stationary phases, XAqua C3 and XAqua C18. An XAqua C3 prep column was used to separate the sample in the first-dimensional preparation, and 14 g of sample was fractionated into eight fractions with a recovery of 82%. An XAqua C18 prep column was selected to prepare high-purity compounds in the second-dimensional preparation for its good orthogonality with the XAqua C3 stationary phase. As a result, major compounds in the sample were isolated with more than 99% purity. This method is a potent method to realize the efficient purification of compounds with high purity from the stem of L. japonica Thunb and it shows great potential in the separation of high-purity compounds from complex samples.

Column chips were fabricated on a silicon wafer with an etched rectangular channel 100 μm (width) × 250 μm (depth) using a deep reactive ion etching (DRIE) process. The column chip for the first GC dimension was 3 m long and was coated with polydimethylsiloxane (DB-1) as the stationary phase. The columns on the second dimensional chip were etched with the same width and depth as the first chip, but the flow channel was split into three parallel columns, 1 m long, on the same sized silicon chip (i.e., 3 cm × 3 cm). These three parallel columns on the second chip were coated with polyethylene oxide (DB-Wax), trifluoropropylpolymethylsilicone (OV-210) and cyanopropylmethylphenylmethylpolysilicone (OV-225), accordingly, in order to provide diversified chromatographic retention. These two chips were connected via a stop-flow configuration to simultaneously generate multiple two-dimensional gas chromatograms for every analysis. This stop-flow μGC × μGCs design allowed the first column to function as a pre-separator and as a sequencing injector for the second parallel-separation chip. Fifteen volatile organic compounds with boiling points that ranged from 80-131 °C with various functional groups were tested using this μGC × μGCs module. Three discrete 2-D chromatograms were generated simultaneously, which demonstrated the advantages of simultaneously combining GC × GC with parallel separation GCs in microchip chromatography. The total traveling length in the column was only 4 m for each eluted peak and fully resolved separation was achieved through the cross reference among triplet 2-D chromatograms.
