- Hadžidedić S1, Uzunović A, Sehić Jazić S, Kocova El-Arini S.Author information 1Agency for Medicinal Products and Medical Devices of B&H , Sarajevo , Bosnia and Herzegovina .AbstractThe aim of this study was to compare the specific characteristics of the S-enantiomer and the racemate of amlodipine besylate (AB) in order to design a robust and stable formulation of the active S-enantiomer which will guarantee continuous performance of the unichiral version of amlodipine. Preformulation studies showed that the S-enantiomer and the racemate exhibit different crystal morphology, particle size distribution and higroscopicity. The S-enantiomer exhibited significantly lower melting temperature compared to the racemate which was in accordance with its higher water solubility and its increased intrinsic dissolution rate. The thermograms of S-amlodipine besylate indicated that dehydration and melting occur at almost the same time and the dehydration event overlaps with the melting peak. Forced degradation tests conducted on both substances showed high levels of degradation into amlodipine related substance D as well as other impurities. Tablets prepared with S-AB, simulating originator's formulation, failed in stability tests due to drug incompatibility with calcium hydrogen phosphate. Therefore, a tablet formulation based on excipients which were confirmed compatible with S-AB was developed and optimized using full factorial design to obtain a dissolution profile comparative to the brand product. Stability studies conducted at 40 °C/75% relative humidity (RH) confirmed that appearance, drug content and drug release of the optimized tablet formulation remained within the recommended limits.

A novel instrument, the Scanning Electrometer for Electrical Double-layer (SEED) has been developed to measure multiple enzyme reactions on a monolith electrode due to immunospecific binding with a mixture of respective analytes. SEED quantitatively maps the local redox reaction by scanning a laser on the array of enzyme monolayer spots immobilized on the monolith electrode. SEED measures the change in local charge state of the EDL that abruptly changes due to the redox reaction. The measurement spot size defined by the size of the laser beam is ~10µm. The SEED signal is linearly proportional to the local redox current density and analyte concentration. The specificity is close to 100%. The SEED readout is compatible with microfluidics platform where the signal degrades less than 2% due to the poly(dimethyl siloxane) (PDMS) body.

Secreted forms of PrP(C) resulting from PrP(C) α-cleavage (PrPN1) or shedding (shed PrP(C)) display neuroprotective activity in neuronal cultures and in mouse models of Aβ-induced neuronal dysfunction. In vitro, recombinant PrPN1 and PrP inhibit Aβ fibrillization. However, the mechanism by which PrPN1 and shed PrP(C) neutralize Aβ oligomers is unclear, and evidence of such neuroprotective activity in Alzheimer's disease (AD) patients is lacking. Here, we show that PrPN1 association with Aβ causes a conformational change resulting in the formation of amorphous and insoluble aggregates that are not compatible with the assembly of Aβs. Using postmortem brain tissues of AD patients, we were able to coimmunoprecipitate Aβ with PrP(C) molecules and observed a coaggregation of Aβ and PrPN1 in the guanidine-extractable fraction presumably representing insoluble amyloid plaques. Furthermore, PrP(C) α-cleavage is increased in AD brains, and we noticed a significant positive correlation between the levels of α-cleavage and of guanidine-extractable Aβ. These data strongly support the hypothesis that PrP(C) α-cleavage is an endogenous neuroprotective mechanism in AD and support the development of PrP(C)-derived peptides as therapeutic molecules for AD.

The resulting tablets were evaluated for the effect of the extent of crosslinking on drug release and matrix swelling. Increase in the concentration of Ca(2+) ions increased the viscosity of gel layer and reduced the water penetration velocity into the matrix with subsequent decrease in swelling of the tablets and drug release. Beyond a certain concentration of Ca(2+) ions, the viscosity of the gel layer decreased and the drug release rate increased primarily due to erosion of the matrix. The mechanism of drug release appeared to be non-Fickian or anomalous transport. The release data also best fitted in zero order equation. The model drug, metronidazole, was compatible with the matrix materials as evident from instrumental analyses. Such formulation may provide flexibility in achieving the desired drug release rate from crosslinked matrix tablets.
