Electronic address: pghe@chem.ecnu.edu.cn.AbstractAn ultrasensitive label-free electrochemiluminescence (ECL) aptasensor for the detection of thrombin was developed based on the specific recognition between tris(bipyridine)ruthenium(II)-β-cyclodextrin (tris(bpyRu)-β-CD) and the anti-thrombin aptamer (aptamer). The NH2-aptamer was first immobilized on the activated glassy carbon electrode (GCE) by coupling interaction. By use of the specific recognition between tris(bpyRu)-β-CD and aptamer, tris(bpyRu)-β-CD was then attached on the surface of GCE. Resulting from the outstanding photoactive properties of tris(bpyRu)-β-CD, the fabricated GCE performed strong ECL signal with the coreactant of 2-(dibutylamino)ethanol (DBAE). However, in the presence of thrombin, aptamer-thrombin bioaffinity complexes were formed, which restricted the recognition activities between aptamer and tris(bpyRu)-β-CD. Thus, fewer tris(bpyRu)-β-CD could be attached on the surface of GCE and led to an obvious decrease of ECL signal. Fortunately, the difference of ECL intensity before and after combination with thrombin was logarithmically linear with the concentration of thrombin in a wide range of 10nM-1pM. Meantime, a detection limit of 0.1pM without any other signal labeling or amplifying procedures indicated that the biosensor performed excellent sensitivity, operability and simplicity.

It briefly covers the milestones of qualitative changes in the field of electrochemiluminescent immunosensors; the peculiarities of the electrochemiluminescent immunoassay formats; the basic mechanisms of ECL detection, main features of early and ongoing approaches in electrochemiluminescent immunoassay commercial instruments, and the recent developments in fabrication of solid-state electrochemiluminescent immunosensors. Moreover, systematized data on biomarkers, immunoassay formats, and novel types of electrochemiluminescent label and immobilization support, such as semiconductor nanocrystals, porous noble metals, graphene, TiO2 nanotube arrays, metal-organic composites, multiwall carbon nanotubes, liposomes, photolummonescent carbone nanocrystals are presented as a table. Considerable efforts have also been devoted towards the following two key points: multiplexing analysis (multi-label, and the multianalyte strategies) and integration in microfluidic lab-on-paper devices with capabilities for point-to-care diagnostics. An immuno-like electrochemiluminescent sensor (based on synthetic receptors-molecularly imprinted polymers), as a new alternative to traditional electrochemiluminescent immunoassay is highlighted. Future perspectives and possible challenges in this rapidly developing area are also discussed.

First, 3-aminobenzeneboronic acid (APBA) was used as the functional monomer and cross-linking reagent, which was polymerized on the surface of silicate-coated magnetic iron oxide nanoparticles (Fe3O4@SiO2 NPs) in the presence of human immunoglobulin G (HIgG), as the template exhibiting the same Fc region but different Fab region to anti-HIV-1 after the addition of the initiator, ammonium persulfate. This process resulted in grafting a hydrophilic molecularly imprinted polymer (MIP) film on the Fe3O4@SiO2 NPs. Thus, MMIPs, which could be reused after eluting the template, were used to recognize and enrich ultra-trace levels of anti-HIV-1. Subsequently, a novel sandwich ECL immunosensor was formed through the immunoreaction between MMIPs conjugated with varied concentrations of anti-HIV-1 and HRP-HIV-1. By the catalysis of HRP immobilized onto HRP-HIV-1 on the ECL system of Luminol-H2O2, a linear response range of the anti-HIV-1 dilution ratio (standard positive serum) was achieved from 1:20,000 to 1:50, with a detection limit of 1:60,000 (S/N=3). The developed method provides a low-cost, simple, and sensitive way for the early diagnosis of HIV infected patients.
