A Master Mariner or MM is the professional qualification required for someone to serve as the person in charge or person in command of a commercial vessel. In England,[where?] the term Master Mariner has been in use at least since the 13th century, reflecting the fact that in guild or livery company terms, such a person was a master craftsman in this specific profession (e.g. Master Carpenter, Master Blacksmith, etc. [2]). In the United States, the term was introduced in the mid-19th century.

United Kingdom
In the UK, the term “Master Mariner” is reserved only for those who hold an unlimited master's certificate. The term “unlimited” (often referred to as unrestricted) indicates that there are no limits on size, power or geographic location on the certificate. It is the highest level of professional qualification amongst mariners and Deck Officers in the British Merchant Navy. The Extra Master's qualification, which was discontinued in the 1990s, used to be the highest professional qualification and it was the pinnacle for any mariner to achieve. There are also various other levels of Master's certificates, which may be restricted or limited to Home Trade/Near Coastal Voyages and/or by gross tonnage. The holder of a restricted Master's certificate is not referred to as a "Master Mariner".
In the British Merchant Navy a Master Mariner who has sailed in command of an oceangoing merchant ship will be titled Captain.[citation needed] A professional seafarer who holds a restricted or limited Master's certificate who has sailed in command of a ship (i.e. appropriate to the size, power or geographic limits of their certificate) can also be titled Captain.
An unrestricted master's certificate is colloquially called a "Master's Ticket" or a "Master's." It is sometimes still referred to as a “Class 1” or “Master Foreign-Going” as it was named during the latter part of the 20th century. The conventions or acts governing the certificate have evolved alongside the shipping industry and the official name of a Master Mariner’s qualification has varied over the years. The UK Maritime & Coastguard Agency, in-line with the amended STCW convention,[1] currently[when?] title the certificate Master Unlimited.[2]
United States
A Master Mariner license comes rated for the maximum gross tonnage of vessel that license-holder is allowed to operate with the highest being a license for "any gross tonnage", colloquially referred to as "unlimited". Additionally, the licenses come with restrictions as to what waters the officer is allowed to operate on (inland, coastal, oceans). An unlimited tonnage ocean license is the highest level license attainable.[3] A Master Mariner who has sailed in command of a commercial vessel is given the title and the appellative of Captain. Also, in common usage anyone who has achieved any USCG master's license commonly refers to themselves as "Captain", regardless of limitations on that license or lack of commercial or command experience.

In order to identify factors that control the production, activity, or secretion of this essential virulence factor, we constructed a Himar1 mariner transposon delivery system and screened 50,000 mutants for a hypohemolytic phenotype on blood agar plates. Approximately 200 hypohemolytic mutants were identified, and the 51 most prominent mutants were screened ex vivo for intracellular growth defects. Eight mutants with a phenotype were identified, and they contained insertions in the following genes: lmo0964 (similar to yjbH), lmo1268 (clpX), lmo1401 (similar to ymdB), lmo1575 (similar to ytqI), lmo1695 (mprF), lmo1821 (similar to prpC), lmo2219 (prsA2), and lmo2460 (similar to cggR). Some of these genes are involved in previously unexplored areas of research with L. monocytogenes: the genes yjbH and clpX regulate the disulfide stress response in Bacillus subtilis, and the prpC phosphatase has been implicated in virulence in other gram-positive pathogens. Here we demonstrate that prsA2, an extracytoplasmic peptidyl-prolyl cis/trans isomerase, is critical for virulence and contributes to the folding of LLO and to the activity of another virulence factor, the broad-range phospholipase C (PC-PLC). Furthermore, although it has been shown that prsA2 expression is linked to PrfA, the master virulence transcription factor in L. monocytogenes pathogenesis, we demonstrate that prsA2 is not directly controlled by PrfA. Finally, we show that PrsA2 is involved in flagellum-based motility, indicating that this factor likely serves a broad physiological role.

The population of pea Stowaway MITEs consists of 103-104 copies dispersed in the genome. Judging from a sequence analysis of 17 isolated Stowaway elements and their flanking genomic regions, the elements are relatively uniform in size and sequence and occur in the vicinity of genes as well as within repetitive sequences. Insertional polymorphism of several elements was detected among various Pisum accessions, suggesting they were still transpositionally active during diversification of these taxa. The identification of several Mariner-like elements (MLEs) harboring intact open reading frames, capable of encoding a transposase, further supports a recent mobilization of the Stowaway elements. Using transposase-coding sequences as a hybridization probe, we estimated that there are about 50 MLE sequences in the pea genome. Among the 5 elements sequenced, 3 distinct subfamilies showing mutual similarities within their transposase-coding regions, but otherwise diverged in sequence, were distinguished and designated as Psmar-1 to Psmar-3. The terminal inverted repeats (TIRs) of these MLE subfamilies differed in their homology to the TIRs of Stowaway MITEs. The homlogy ranged from 9 bp in Psmar-3 to 30 bp in Psmar-1, which corresponds to the complete Stowaway TIR sequence. Based on this feature, the Psmar-1 elements are believed to be the most likely candidates for the master elements of the Stowaway MITEs in pea.
