Pseudomonas strains were tested against the causal agents of tomatoes damping-off (Sclerotinia sclerotiorum), root rot (Fusarium solani), and causal agents of stem canker and leaf blight (Alternaria alternata). For this purpose, dual culture antagonism assays were carried out on 25% tryptic soy agar, King B medium and potato dextrose agar to determine the effect of the strains on mycelial growth of the pathogens. In addition, strains were screened for their ability to produce exoenzymes and siderophores. All the strains significantly inhibited Alternaria alternata, particularly in 25% TSA medium. Antagonistic effect on Sclerotinia sclerotiorum and Fusarium solani was greater on King B medium. Protease was produced by 30% of the strains, but no strain produced cellulase or chitinase. Finally, the selected Pseudomonas strain, Psf5, was evaluated on tomato seedling development and as a potential candidate for controlling tomato damping-off caused by Sclerotinia sclerotiorum, under growth chamber conditions. In vivo studies resulted in significant increases in plant stand as well as in root dry weight. Psf5 was able to establish and survive in tomato plants rhizosphere after 40days following the planting of bacterized seeds.

Specialized metabolites such as phenolics are considered to be an important component of plant defense, including SIR, but the antimicrobial activity of many of these putative defensive chemicals remains untested at realistic concentrations and in conjunction with each other. Here, we examined the anti-Diplodia activity of several previously identified Austrian pine phenolics associated with SIR by comparing the diameters of fungal colonies grown on media amended with ferulic acid, coumaric acid, taxifolin, pinosylvin, pinosylvin monomethyl ether and lignin. All of the compounds were tested both individually and as clusters (combinations) previously determined to occur in planta in a co-regulated fashion. Both the individual compounds and clusters were tested at constitutive concentrations and pathogen-induced concentrations linked to an SIR phenotype. Lignin possessed the strongest antifungal activity individually, and clusters at the SIR concentrations had the greatest antifungal effects, achieving fungistasis. This study exemplifies the value of evaluating potential biomarkers of resistance at in planta concentrations that are associated with the systemically resistant phenotype, and provides strong evidence that co-regulation of chemical defenses potentiates such a phenotype.

In the present field study, we recovered Aphelenchoides spp. in almost all (97.0 %) of 133 blight canker tissue assays (three 5-g samples each) from four eastern states. High mean population densities (227 to 474 nematodes per 5 g tissue) of Aphelenchoides spp. were recovered from cankers in Virginia, West Virginia, and Tennessee but not from New Hampshire (mean = 75 nematodes per 5 g tissue). Overall, most canker assays yielded population densities less than 200 nematodes per 5 g tissue. All of 12 very small or young cankers yielded a few to many Aphelenchoides spp. Regression analysis indicated greatest recovery of Aphelenchoides spp. occurred in the month of May (r = 0.94). The results indicate that Aphelenchoides spp. appear to be widespread in blight cankers on American chestnut trees and could play a role in biocontrol of chestnut blight.
