China.AbstractFamilial hypercholesterolemia (FH) is a genetic disorder characterized by a high serum concentration of low‑density lipoprotein (LDL) cholesterol. The high LDL cholesterol level leads to an excess deposition of cholesterol in the arterial walls and accelerated atherosclerosis, thereby increasing the risk of premature coronary heart disease. In the present study, we used a DNA microarray approach to identify gene expression profiles that distinguish patients with FH from healthy control subjects. Furthermore, transcription factors (TFs), microRNAs (miRNAs), target genes and pathways were analyzed to explore the potential transcriptional interactions occurring in FH. Publicly available microarray and regulation data were used to construct a regulatory network to identify additional genes related to FH and their interactions. The results revealed that specificity protein 1 (SP1), signal transducer and activator of transcription 1 (STAT1) and spleen focus forming virus (SFFV) proviral integration oncogene spi1 (SPI1) play a central role in the FH regulatory network. In addition, the TF, upstream transcription factor 2, c-fos interacting (USF2) and the gene, Wiskott-Aldrich syndrome (WAS), were identified to be associated with FH, although no reports for these proteins exist in the literature. Overall, transcriptional network analysis proved to be effective approach to identify novel targets for FH therapy.

To investigate the possible role of Wnt signaling in the regulation of ovarian follicular development, secondary follicles were isolated and cultured in vitro in the presence or absence of its activator (LiCl or Wnt3a) or inhibitor (IWR-1). We have demonstrated that activation of β-catenin signals by activators dramatically suppressed follicular development by increasing granulosa cell apoptosis and inhibiting follicle steroidogenesis. In contrast, inhibition of Wnt signaling by IWR-1 was observed with better developed follicles and increased steroidogenesis. Further studies have shown that the transcription factor Forkhead box O3a (Foxo3a) and its downstream target molecules were modulated by the activators or the inhibitor. These findings provide evidence that Wnt signaling might negatively regulate follicular development potentially through Foxo3a signaling components.

Its surface can be engineered to meet specific needs for technical, medical or materials applications. However, not all technically desirable TMV coat protein (CP) mutants can be propagated in plants successfully, if they change the efficiency of virion assembly. In order to circumvent this problem, a novel wild type (wt) CP-assisted and RNA-directed assembly procedure was designed for a recalcitrant CP mutant: Although pure hexahistidine-tagged CP cannot form particles on its own with TMV RNA in vitro, it was integrated into full-length particles if blended with wt CP in different proportions. The resulting rods formed dense monolayers with short range alignment on silicon substrates, substantially different from the largely wavy patterns obtained with wt TMV. Since they also mediated efficient ZnO deposition under mild conditions, the approach has yielded a new class of biotemplates which are amenable to the formation of nanostructured hybrid materials with adjustable texture for various applications.
