- ISOLATION, SCREENING AND PRODUCTION STUDIES OF URICASE PRODUCING BACTERIA FROM POULTRY SOURCES.
- Nanda P1, Babu PE.Author information 1a Department of Chemical Engineering , National Institute of Technology Karnataka, Srinivasnagar , Surathkal , Karnataka , 575 025 , India.AbstractUricase (Urate oxidase E.C.1.7.3.3) is a therapeutic enzyme, which is widely used to catalyze the enzymatic oxidation of uric acid in the treatment of hyperuricemia and gout diseases. In this present study, three bacterial species capable of producing extracellular uricase were isolated from poultry source and screened based on the size of the clear zone using uric acid agar plate. Bacterial species capable of producing uricase with highest uricolytic activity was identified as Bacillus cereus strain DL3 using 16SrRNA gene sequencing approach. The time course study of uricase production was performed and the medium was optimized. Carboxy methylcellulose and asparagine were found to be the best carbon and nitrogen sources. Maximum uricolytic activity was observed at pH of 7.0 with an inducer concentration of 2.0 g/L. Inoculum size of 5--% gave maximum uricolytic activity. The maximum uricolytic activity of 15.43 U/mL was achieved at optimized conditions, which is 1.61 times more than the initial activity. Further, enzymatic stability was determined at different pH and temperature.

- Spray drying formulation of albendazole microspheres by experimental design. In vitro-in vivo studies.
- García A1, Leonardi D, Piccirilli GN, Mamprin ME, Olivieri AC, Lamas MC.Author information 1IQUIR - CONICET .AbstractAbstract Both an experimental design and optimization techniques were carried out for the development of chitosan-pectin-carboxymethylcellulose microspheres to improve the oral absorption of albendazole as a model drug. The effect of three different factors (chitosan, pectin and carboxy methyl cellulose concentrations) was studied on five responses: yield, morphology, dissolution rate at 30 and 60 min, and encapsulation efficiency of the microspheres. During the screening phase, the factors were evaluated in order to identify those which exert a significant effect. Simultaneous multiple response optimizations were then used to find out experimental conditions where the system shows the most adequate results. The optimal conditions were found to be: chitosan concentration, 1.00% w/v, pectin concentration 0.10% w/v and carboxymethylcellulose concentration 0.20% w/v. The bioavailability of the loaded drug in the optimized microspheres was evaluated in Wistar rats which showed an area under curve (AUC) almost 10 times higher than the pure drug.
