Electronic address: prbueno@iq.unesp.br.AbstractThe transducer faradaic signals of molecularly receptive interfaces associated with specific target binding can be sensitively monitored by electrochemical impedance and/or capacitance spectroscopies. A comparative evaluation of both impedimetric (associated with charge transfer resistance) and capacitive (associated with faradaic density of states) approaches was undertaken using C-reactive protein (CRP) antigen and antibody interaction as biomolecular binding model. Aiming at constructing redox free (impedimetric) and redox tethered receptive (capacitive) interfaces engineered by self-assembly monolayer, CRP sensitivity and limit of detections were comparatively assessed regarding biosensor capabilities. Binding affinity constant between CRP and anti-CRP interfacial receptor sites were additionally evaluated by the Langmuir adsorption model. Both the impedimetric and capacitive approaches reported similar values of experimental analytical parameters albeit the latter was found to have the advantage of requiring no solution redox reporter thus making it highly suitable for use in multiplexing affinity arrays.

The changes in surface properties of the parylene-N film after plasma-treatment were analyzed using contact angle microscopy and AFM. To demonstrate the high protein-immobilization efficiency of the plasma-treated parylene-N film, the immobilization efficiencies of differently modified surfaces were compared using model proteins with different surface charges, such as streptavidin (pI=5, negatively charged at pH 7), horseradish peroxidase (pI=6.6, nearly neutral at pH 7), and avidin (pI=10, positively charged at pH 7). The application of the plasma-treated parylene-N film as an SPR biosensor was also tested by immobilizing model proteins. An SPR biosensor based on the plasma-treated parylene-N film was developed for the detection of the human hepatitis virus surface antigen (HBsAg), and the plasma-treated parylene-N film was estimated to improve the sensitivity of SPR biosensor as much as 1000-fold by enhancing immobilization of receptor antibodies.

Lin(-)MHCII(+)CD11c(+)CD205(+) mDCs (0.2% blood mononuclear cells) isolated from bovine blood were heterogeneous in cell size and CD205 expression. Using highspeed cell sorting, Lin(-)MHCII(+)CD11c(+)CD205(+) DCs were sorted into CD205(Hi) and CD205(Lo) subpopulations which were phenotypically distinct and differed significantly (P<0.01) in TLR gene expression. CD205(Hi) and CD205(Lo) mDCs were more efficient in macropinocytosis than monocytes and expressed no or little detectable non-specific esterase activity. CD205(Lo) mDCs efficiently activated purified allogeneic T cells and the addition of TLR agonists did not significantly alter this antigen presentation capacity. T cell activation by CD205(Lo) mDCs was associated with differential up-regulation of CD40, CD80, CD86 and TGFβ1 gene expression when compared to CD205(Hi) mDCs. In conclusion, two phenotypically and functionally distinct CD11c(+)CD205(+) mDCs were isolated from blood that had an equal capacity to acquire antigen but markedly different capacities to activate T cells.
