In this approach, information about the action of GTs (consisting of possible sugar residues used as donors and/or acceptors, as well as the anomeric configuration and/or substitution position in the respective glycosidic linkages) is extracted from the Escherichia coli O-antigen database and is submitted, together with the unassigned NMR data, to the CASPER program. This time saving methodology, which alleviates the need for chemical analysis, was successfully implemented in the structural elucidation of the O-antigen PS of E. coli O59. The repeating unit of the O-specific chain was determined using the O-deacylated PS and has a branched structure, namely, →6)[α-d-GalpA3Ac/4Ac-(1 → 3)]-α-d-Manp-(1 → 3)-α-d-Manp-(1 → 3)-β-d-Manp-(1 → 3)-α-d-GlcpNAc-(1→. The identification of the O-acetylation positions was efficiently performed by comparison of the 1H,13C HSQC NMR spectra of the O-deacylated lipopolysaccharide and the lipid-free PS in conjunction with chemical shift predictions made by the CASPER program. The side-chain d-GalpA residue carries one equivalent of O-acetyl groups at the O-3 and O-4 positions distributed in the LPS in a 3:7 ratio, respectively. The presence of O-acetyl groups in the repeating unit of the E. coli O59 PS is consistent with the previously proposed acetyltransferase WclD in the O-antigen gene cluster.

This article analyses the available biological, physico-chemical, and nucleotide sequence evidence that seems to indicate that OFV and several other Brevipalpus mite-transmitted short bacilliform (-)ssRNA viruses are likely related and may be classified taxonomically in novel species in a new free-floating genus Dichorhavirus.

Electronic address: picariello@isa.cnr.it.AbstractFor the purpose of a varietal assessment, the berry skin anthocyanin profiles of 11 ancient native red grape varieties, sampled within the Irpinian area (Southern Italy), were compared to those of three reference Vitis vinifera cultivars and of a Kober 5BB rootstook hybrid (Vitisberlandieri×Vitisriparia). The 3,5-O-diglucoside anthocyanins and their acylated derivatives were monitored as signature compounds of non-V. vinifera grapes, using both reversed phase-high performance liquid chromatography (RP-HPLC) and matrix assisted laser desorption ionisation-time of flight (MALDI-TOF) mass spectrometry (MS). One variety (i.e. Tenta) was demonstrated to be an interspecific hybrid cross. Three additional varieties, namely Lacrima Nera, Aglianicone and a yet-unnamed variety, were classified as "late generation hybrids" (or non-V. vinifera×V. vinifera hybrids) because of a very diluted hybrid character, that was revealed only by MS methods. Five cultivars, i.e. Aglianico Lasco, Cannella, Coda di Volpe Rossa, Mentuonico, Olivella Nera, were catalogued as purebred V. vinifera. Due to the peculiar anthocyanin profile one variety (Tuccanese) remained unassigned. The methodology is of general applicability to support the process of varietal discrimination on a molecular basis with the objective of classifying autochthonous old grapevine varieties.
