The terminal web is a filamentous structure found at the apical surface of epithelial cells that possess microvilli. It is composed primarily of actin filaments stabilized by spectrin, which also anchors the terminal web to the apical cell membrane. The presence of myosin II and tropomyosin helps to explain the contractile ability of the terminal web. When contracted, the terminal web causes a decrease in diameter of the apex of the cell, causing the microvilli, which are anchored into the terminal web through their stiff actin fibers, to spread apart. This spreading apart of the microvilli aids cells in absorption.[1][2][3][4]

- Structural and biochemical characterisation of the KLHL3-WNK kinase interaction important in blood pressure regulation.
- Schumacher FR, Sorrell FJ, Alessi DR, Bullock AN, Kurz T.AbstractWith no lysine (K) kinase-1 (WNK1) and WNK4 regulate blood pressure by controlling the activity of ion co-transporters in the kidney. Groundbreaking work has revealed that the ubiquitylation and hence levels of WNK isoforms are controlled by a Cullin-RING E3 ubiquitin ligase complex (CRL3KLHL3) that utilises Cullin3 (CUL3) and its substrate adaptor, kelch-like protein 3 (KLHL3). Loss of function mutations in either CUL3 or KLHL3 cause the hereditary high blood pressure disease Gordon's syndrome by stabilising WNK isoforms. KLHL3 binds to a highly conserved degron motif located within the C-terminal non-catalytic domain of WNK isoforms. This interaction is essential for ubiquitylation by CRL3KLHL3 and disease-causing mutations in WNK4 and KLHL3 exert their effects on blood pressure by disrupting this interaction. In the present study we report on the crystal structure of the KLHL3 kelch domain in complex with the WNK4 degron motif. This reveals an intricate web of interactions between conserved residues on the surface of the kelch domain β-propeller and the WNK4 degron motif. Importantly, many of the disease-causing mutations inhibit binding by disrupting critical interface contacts. We also present the structure of the WNK4 degron motif in complex with the kelch-like 2 (KLHL2) protein that has also been reported to bind WNK4. This confirms that KLHL2 interacts with WNK kinases in a similar manner to KLHL3, but strikingly different to how another kelch-like protein KEAP1 binds to its substrate NRF2. This study provides further insights into how kelch-like adaptor proteins recognise their substrates and provides a structural basis for how mutations in WNK4 and KLHL3 lead to hypertension.

The exact nature of how these compounds have an inhibitory effect on HCV is unknown; however, major resistance mutations appear in the N-terminal region of NS5A that include the amphipathic helix and domain 1. The dimeric symmetry of these compounds suggests that they act on a dimer of NS5A, which is also consistent with the presence of dimers in crystals of NS5A domain 1 from genotype 1b. Genotype 1a HCV is less potently affected by these compounds and resistance mutations have a greater effect than in the 1b genotypes. We have obtained crystals of domain 1 of the important 1a NS5A homologue and intriguingly, our X-ray crystal structure reveals two new dimeric forms of this domain. Furthermore, the high solvent content (75%) makes it ideal for ligand-soaking. Daclatasvir (DCV) shows 2-fold symmetry suggesting NS5A dimers may be of physiological importance and serve as potential binding sites for DCV. These dimers also allow for new conformations of a NS5A expansive network which could explain its operation on the membranous web. Additionally, sulfates bound in the crystal structure may provide evidence for the previously proposed RNA binding groove, or explain regulation of NS5A domain 2 and 3 function and phosphorylation by domain 1.
