We evaluated the cytotoxicity of a series of pristine and organo-modified nanoclays in different cell lines. The calculated IC50 values for cell viability ranged from 1.4 to 47 µg/mL for the six organoclays used and were above 100 µg/mL for the pristine nanoclays. The IC50 values of the organoclays were driven by the proportion and structure of the quaternary ammonium compound used as surface organic modifier. No differences in cell toxicity were observed between the large and small-sized (additional milling step) nanoclay batches, although their size differences related mostly to upper range of the size distribution. Despite their lower toxicity, pristine nanoclays induced apoptosis and were found in cytoplasmic vesicles of exposed cells. Organoclays were also found in cytoplasmic vesicles, although the size of the agglomerates was larger and the efficiency of uptake was considerably lower.

A particular miRNA can have hundreds of targets; therefore, miRNAs cumulatively influence the expression of a large proportion of genes. The functions of miRNAs in human diseases have been studied since their discovery in mammalian cells approximately 12 years ago. However, the role of miRNAs in allergic disease has only very recently begun to be uncovered. The purpose of this review is to provide an overview of the functions of miRNAs involved in the development of allergic diseases. We describe here the functions of miRNAs that regulate Th2 polarization and influence general inflammatory and tissue responses. In addition, we will highlight findings about the functions of extracellular miRNAs as possible noninvasive biomarkers of diseases with heterogeneous phenotypes and complex mechanisms and briefly discuss advances in the development of miRNA-based therapeutics.

Screening low-risk individuals for coronary artery disease.

Efforts to improve early detection of coronary artery disease among low-risk individuals, or to improve risk assessment, might be justified by this large population burden. The most promising tests for improving risk assessment, or early detection, include the coronary artery calcium (CAC) score, the ankle-brachial index (ABI), and the high-sensitivity C-reactive protein (hsCRP). Data regarding the role of additional testing in low-risk populations to improve early detection or to enhance risk assessment are sparse but suggest that CAC and ABI may be helpful for improving risk classification and detecting the higher-risk people from among those at lower risk. However, in the absence of clinical trials in this patient population, such as has recently been proposed, we do not recommend routine use of any additional testing or screening in low-risk individuals at this time.
