- Lanthanide-based nanocrystals as dual-modal probes for SPECT and X-ray CT imaging.

Herein, small size PEG-EuOF:(153)Sm nanocrystals (∼5 nm) (PEG = poly(ethylene glycol)bis(carboxymethyl)ether) combined with the radioactive and X-ray absorption properties were synthesized. The distribution of the PEG-EuOF nanocrystals in living animals was studied by ex vivo radioassay, inductively coupled plasma-atomic emission spectrum (ICP-AES) analysis and in vivo SPECT imaging, which indicated that the small size PEG-EuOF:(153)Sm had long blood retention time (blood half-life (t1/2) reach to 4.65 h) and were eliminated significantly through biliary/gastrointestinal pathway in vivo. Meanwhile, benefiting from the high attenuation ability of Eu, the small size PEG-EuOF was successfully applied for lymph node CT imaging, extending the bio-applications of these small nanocrystals. The results of cytotoxicity and in vivo toxicity also showed that the PEG-EuOF nanocrystals have relatively low toxicity, which suggest their safety for in vivo imaging. The studies provide preliminary validation for the use of PEG-EuOF nanocrystals for in vivo bioimaging applications.

- Computed tomography of Lipiodol-loaded biodegradable pasty polymer for implant visualization.
- Sosna J1, Havivi E, Khan W, Appelbaum L, Nyska A, Domb AJ.Author information 1Department of Radiology, Hadassah - Hebrew University Medical Center, Jerusalem, Israel; Department of Radiology, Beth Israel Deaconess Medical Center, Harvard Medical School, Boston, MA, USA.AbstractTargeted delivery of drug-loaded implants for regional drug therapy has become an important approach to therapy. Simple and reproducible imaging methodologies to evaluate the implant noninvasively are needed. The goal of this work was to noninvasively evaluate the visibility, shape and degradation of a biodegradable implant containing Lipiodol (an X-ray contrast medium) by computed tomography (CT). For in vitro evaluation, Lipiodol was incorporated in poly(sebacic-co-ricinoleic acid) [P(SA:RA)], a biodegradable injectable pasty polymer, and CT visibility was assessed. For ex vivo evaluation, bovine liver was injected with the polymer-loaded Lipiodol; for in vivo evaluation rats were injected subcutaneously with Lipiodol in polymer and CT was performed. We show that polymer diameter at CT correlates with implant weight and pathological measurements. Polymer formulation containing 5% Lipiodol was visible on CT in vitro. Ex vivo tests showed a round polymer deposit at the injection site compared with free dispersion of Lipiodol alone. Correlation between implant size at CT scan and surgery at 48 h was R(2) = 0.78. Average CT diameter at 9 days was 14.2 ± 2.8 mm in rats injected with Lipiodol in the polymer formulation, as compared with 7.3 ± 1.1 mm in controls. After 9 days, the implant degraded into several zones containing inflammatory cells seen on CT as areas with increased heterogeneity. In conclusion, Lipiodol incorporated in P(SA:RA) is visible on CT, and polymer degradation can potentially be monitored noninvasively. This method can be widely applied to follow changes in biodegradable implants. Copyright © 2014 John Wiley & Sons, Ltd.

- Validation of CT doses of SPECT/CT and PET/CT hybrid devices: lessons learned.
- Sera T1, Porubszky T, Papos M, Elek R, Besenyi Z, Gion K, Bartha A, Pellet S, Pavics L.Author information 1aDepartment of Nuclear Medicine, University of Szeged bEuromedic Diagnostics, Szeged c'Frédéric Joliot-Curie' National Research Institute for Radiobiology and Radiohygiene, Budapest, Hungary.AbstractThe aim of the study was to check the validity of computed tomographic (CT) doses exhibited by SPECT/CT and PET/CT hybrid devices. Dose measurements were taken from four SPECT/CT and four PET/CT cameras commercially available from different manufacturers. A calibrated ionization chamber was placed in whole-body or head phantoms for the acquisition of CT images with clinically used parameters. Computed tomography dose index (CTDIvol) values were calculated according to the IEC 60601-2-44:1999 formula. The measured CTDIvol doses were compared with those preprogrammed by the manufacturer. In the case of the whole-body phantom, the differences between the measured and displayed values varied between -31 and +24% [European document RP162 (2012) sets up the limit for acceptance criterion as ±20%]. The head phantom data showed either an agreement between -10 and +24%, or an underestimation by two-fold. The latter seemed to be because, while preprogramming the doses, the manufacturer had used the whole-body phantom instead of a proper head phantom. The results of the work demonstrate the need for individual dosimetric calibration of every single X-ray tube. Dosimetric checks should be included in the regular quality control programmes of the SPECT/CT and PET/CT devices. Special attention should be paid to head-and-neck and paediatric protocols, in which the use of a head phantom is recommended for dose calibration.
