Response in the hindfoot pad and popliteal lymph node of C57BL mice to infection with Mycobacterium marinum.

Mor N, Lutsky I, Levy L.AbstractC57BL mice inoculated in the hindfoot pads with 5 X 10(3) viable Mycobacterium marinum developed a localized disease process, characterized by swelling of the foot and increases in the number of acid-fast bacilli and colony forming units recovered. These changes became maximal 10 to 14 days after inoculation and then decreased in intensity. An acute inflammatory response appeared in the hindfoot pad during the first 24 h and increased in intensity during the next few days. By seven days after inoculation, the polymorphonuclear leukocytes had been largely replaced by lymphocytes and macrophages which led, during the next two weeks, to the extensive formation of granulomas. Epithelioid granulomas developed after at least 90% of the organisms had been killed. Simultaneously the popliteal lymph node increased greatly in size due to hyperplasia of the paracortical area, which contained a large number of pyroninophilic cells, and packing of sinusoids with small lymphocytes. Well-formed epithelioid granulomas containing acid-fast bacilli developed in the popliteal node. A small number of viable bacilli were found in both the hindfoot pad and the popliteal node 18 months after inoculation; this was accompanied by solid resistance to secondary challenge.

Dynamic and morphological responses of draining lymph nodes to single and repeated applications of oxazolone to guinea-pig skin.

Myking AO.AbstractAuricular lymph nodes in guinea-pigs were studied for 3 weeks under continuous stimulation with oxazolone applied to the ear skin. Quantitation of 3H-Tdr labelled paracortical lymphocytes following pulse labeling, demonstrated a marked, but only transitory rise in the proportion of cells in DNA synthesis. In spite of this, the total number of cells in S-phase continued to rise during the remaining part of the observation time, as a result of a steady increase of the paracortical cell mass. The variation in the proportion of large, pyroninophilic blast cells, revealed a pattern similar to that of the labelling index. A high proportion of blast cells was found only in the initial phase of the reaction, while the chronic response was characterized by a low proportion, no different from the starting level. Paracortical enlargement appeared to be the most reliable morphological criterion on which a chronically stimulated paracortex could be distinguished from an "unstimulated". The initial paracortical response, similar to a primary reaction, was followed by an equally pronounced development of germinal centres and plasma cells. These were also persistent features under continued stimulation. The reported changes are most likely specific responses to oxazolone stimulation.

Effects of estrogen on the lymphoid regeneration and immune response in irradiated and marrow-reconstituted mice.

Kotani M, Fujii H, Tsuchiya H, Matsuno K, Ekino S, Harada S.AbstractVarious doses of estriol (E3) were given to mice intraperitoneally, immediately after lethal irradiation and marrow reconstitution. The assessment of the plaque-forming cell (PFC) response to sheep erythrocytes in the spleen and the histological assessment of lymphoid tissues were carried out 30 days later. The effects appeared to be dose-dependent and resulted in a marked suppression of the PFC response. The depletion of lymphocytes was dramatic and dose-dependent in the thymus, and in the thymus-dependent and in the thymus independent areas of the peripheral lymphoid tissues. These results suggest that E3 acts on the differentiation of stem or precursor cells toweard both the populations of T and B lymphocytes. Although E3, given on day 7 after irradiation and marrow reconstitution, suppressed the lymphoid regeneration and PFC response markedly, E3 given on day 14 had no effect. On day 7 the majority of regenerating lymphoid tissues were large pyroninophilic cells and on day 14, small lymphocytes. These results suggest that the precursor or immature lymphocytes are sensitive to E3, while mature lymphocytes are resistant. Lymphoid regeneration and PFC response were retarded in mice irradiated and reconstituted with bone marrow cells from donors pretreated with E3. These results suggest that E3 acts on the stem or precursor cells capable to differentiate in the direction of lymphoid populations and reduce their number in the bone marrow.
