Electronic address: zhjl@imm.ac.cn.AbstractThe metabolism of Tartary buckwheat was investigated using a strategy from single bioactive compounds to complex Tartary buckwheat extract. Firstly, the metabolites of different structural compounds were investigated by an in situ liver-intestinal perfusion model and the metabolic pathways were proposed. Furthermore, Tartary buckwheat extract in rats was elucidated on the basis of the metabolism information of single compounds. High-performance liquid chromatography coupled with high resolution mass and multiple-stage mass spectrometry (HPLC-HRMS/MS(n)) was performed to characterise and identify 19 metabolites in perfusate and intestinal content after administration of single compounds to an in situ liver/intestinal perfusion model and 16 metabolites and 6 components in rat faeces, urine, bile, and plasma after oral administration of Tartary buckwheat to rats. Five new metabolites were identified as the glucuronidation and sulfation products of N-trans-feruloyltyramine and the methylation product of quercetin-3-O-[β-d-xyloxyl-(1→2)-α-l-rhamnoside]. The metabolic pathways of phenylpropanoid glycosides and N-trans-feruloyltyramine were proposed for the first time.

Merrill, from China, Japan, and Korea was examined by high-performance liquid chromatography. The profiles of 12 isoflavones identified from the grains were subjected to data-mining processes, including partial least-squares discriminant analysis (PLS-DA), Pearson's correlation analysis, and hierarchical clustering analysis (HCA). Although PLS-DA did not reveal significant differences among extracts of soybean from 3 countries, the results clearly show that the variation between varieties was low. The CS02554 variety was separate from the others in the first 2 principal components of PLS-DA. HCA of these phytochemicals resulted in clusters derived from closely related biochemical pathways. Daidzin, genistin, and glycitin contents were significantly correlated with their respective malonyl glycoside contents. Daidzein content correlated positively with genistein content (r=0.8189, P<0.0001). The CS02554 variety appears to be a good candidate for future breeding programs, as it contains high levels of isoflavone compounds. These results demonstrate the use of metabolite profiling combined with chemometrics as a tool for assessing the quality of food and identifying metabolic links in biological systems.

The contents of flavonoids (by HPLC-DAD) and proanthocyanidins (n-butanol/HCl assay), reducing capacity (ferric ion reducing antioxidant power, FRAP) and anti-glycation activity by a bovine serum albumin (BSA)/fructose model system were analysed in vitro. A liking test was performed with consumers. Stabilization was carried out by either an intensive autoclave treatment or an optimised microwave-treatment achieving 6D-reduction of the target microorganism (Alicylobacillus acidoterrestris). In the fortified tomato purees, the solubility of proanthocyanidins decreased, but was partly restored by autoclave treatment, which also caused deglycosylation of flavonol glycosides. Microwave treatment did not show any effect on phenolics. The reducing capacity and ability to inhibit protein glycation greatly increased in the fortified purees. The particle sizes of solids in the formulations played a major role with respect to the consumers' liking, with the smallest fraction showing maximum ratings.
